Nicotinamide N-methyltransferase (NNMT), a key cytoplasmic protein in the human body, is accountable to catalyze the nicotinamide (NCA) N1 -methylation through S-adenosyl-L-methionine (SAM) as a methyl donor, which has been linked to many diseases. Although extensive studies have concerned about the biological aspect, the detailed mechanism study of the enzyme function, especially in the part of protein dynamics is lacking.

Two modi®cations of 5-acetimidoylamino-7-acetamido-3,5,7,9-tetradeoxy-D-glycero-D-galacto-non-2-ulosonic acid (5-N-acetimidoyl-7-N-acetyllegionaminic acid) in the O-chainpolysaccharide (OPS) of theLegionellapneumophila serogroup 1 lipopolysaccharide (LPS) concern N-methyla-tionof the 5-N-acetimidoyl group in legionaminic acid. Both N-methylated substituents, the (N,N-dimethylacetimidoyl) amino and acetimidoyl(N-methyl)amino group, could be allocated to one single legionaminic acid residue in the long-and middle-chain OPS, respectively. ...

Special oligonucleotides for targeted gene correction have attracted increasing attention recently, one of which is the chimericRNAÆDNAoligonucleotide (RDO) system. RDOs for targeted gene correction were first designed in 1996, and are typically 68 nucleotides in length including continuous RNA andDNA sequences (RNA is 2¢-O-methyl-modified).

EcoRII DNA methyltransferase (M.EcoRII) recognizes the 5¢…CC*T/AGG…3¢ DNA sequence and catalyzes the transfer of the methyl group fromS-adenosyl-L-methionine to the C5 position of the inner cytosine residue (C*). Here, we study themechanismof inhibitionofM.EcoRII byDNA containing 2-pyrimidinone, a cytosine analogue lacking an NH2 group at the C4 position of the pyrimidine ring. Also, DNA containing 2-pyrimidinone was used for probing contacts ofM.EcoRII with functional groups of pyrimidine bases of the recognition sequence....

Human methionine synthase (hMS) is a multidomain cobalamin-dependent enzyme that catalyses the conversion of homocysteine to methionine by methyl group transfer. We report here the 1.6 A˚ crystal structure of the C-terminal activation domain of hMS.

Juvenile hormone controls the timing of insect metamorphosis. As a final step of juvenile hormone biosynthesis, juvenile hormone acid O-methyl-transferase (JHAMT) transfers the methyl group from S-adenosyl-l-methi-onine to the carboxyl group of farnesoic acid and juvenile hormone acid.

The protein arginine methyltransferase (PRMT) family of enzymes cata-lyzes the transfer of methyl groups from S-adenosylmethionine to the gua-nidino nitrogen atom of peptidylarginine to form monomethylarginine or dimethylarginine. We created several less polar analogs of the specific PRMT inhibitor arginine methylation inhibitor-1, and one such compound was found to have improved PRMT inhibitory activity over the parent molecule.

The Dnmt3a DNA methyltransferase is responsible for establishing DNA methylation patterns during mammalian development. We show here that the mouse Dnmt3a DNA methyltransferase is able to transfer the methyl group fromS-adenosyl-L-methionine (AdoMet) to a cysteine residue in its catalytic center. This reaction is irreversible and relatively slow.
