  • Ebook "Advanced time-correlated single photon counting applications" is an attempt to bridge the gap between the instrumental principles of multi-dimensional time-correlated single photon counting (TCSPC) and typical applications of the technique. Written by an originator of the technique and by sucessful users, it covers the basic principles of the technique, its interaction with optical imaging methods and its application to a wide range of experimental tasks in life sciences and clinical research.

  • In clay-rich sedimentary rocks, the pore network is considered to play a key role in controlling transport properties. The pore size distribution (PSD) of clay-rich rocks, the geometrical features of the pore network, and the clay content are parameters governing the diffusion process through clay formations.

  • Rapid improvements in light and electron microscopy imaging techniques and the development of 3D anatomical atlases necessitate new approaches for the visualization and analysis of image data. Pixel-based representations of raw light microscopy data suffer from limitations in the number of channels that can be visualized simultaneously.

  • In recent years, hyperspectral microscopy techniques such as infrared or Raman microscopy have been applied successfully for diagnostic purposes. In many of the corresponding studies, it is common practice to measure one and the same sample under different types of microscopes.

  • Because of its non-destructive nature, label-free imaging is an important strategy for studying biological processes. However, routine microscopic techniques like phase contrast or DIC suffer from shadow-cast artifacts making automatic segmentation challenging.

  • Transmission electron microscopy (TEM) is a microscopic technique in which a beam of electrons is transmitted through an ultra-thin specimen, interacting with the specimen as it passes through. An image is formed by the interaction of the electrons transmitted through the specimen; the image is magnified and focused onto an imaging device, such as a fluorescent screen or on a layer of photographic film, or to be detected by a sensor such as a CCD camera.

  • Analyses of the spatial localization and the functions of bacteria in host plant habitats through in situ identification by immunological and molecular genetic techniques combined with high resolving microscopic tools and 3D-image analysis contributed substantially to a better understanding of the functional interplay of the microbiota in plants. Among the molecular genetic methods, 16S-rRNA genes were of central importance to reconstruct the phylogeny of newly isolated bacteria and to localize them in situ.

  • In this paper we present the construction of a highly sensitive FCS instrument and the measurement results from a sample of semiconductor quantum dots. We provide the analysis procedure for determining the hydrodynamic radius of the quantum dots and compare the results with the ones obtained directly from electron microscope imaging. The good agreement indicates the reliability of the FCS technique and open the way for further applications of this technique in studying nanoparticles.

  • The GNSs were also bioconjugated with anti-HER2 monoclonal antibody for diagnostic breast cancer cells using dark field microscope technique. These GNS NPs play a role as nanoheaters transforming light to heat. With the present of these GNS NPs at volume density of 3.6×1010 NPs.cm3 in chicken tissue samples, illuminated by 808 nm laser at the power density of 62 W.cm2 the temperature of tissue sample reachs 110˚ C after 20 minutes illumination.

  • The invention of scanning tunneling microscope (STM) by Binnig and his colleagues in 1982 opened up the possibility of imaging material surfaces with spatial resolution much superior to the conventional microscopy techniques. The STM is the first instrument capable of directly obtaining three-dimensional images of solid surfaces with atomic resolution. Even though STM is capable of achieving atomic resolution, it can only be used on electrical conductors. This limitation has led to the invention of atomic force microscope (AFM) by Binnig and his co-workers in 1986.

  • It is no surprise to see the micro-Raman Group at Lille come forth with this timely publication to document the present state of Raman microscopy. A quarter century has passed since the early attempts at Raman microsampling when the field began to merge with, and complement, other microprobe techniques. In the late 1960s to the early '70s, it was mainly the electron beam methods that opened up the microscopic domain to instrumental analysis, aside from classical light microscopy.

  • In 1665, a book was published that inaugurated the use of the microscope to investigate the natural world. The author was Robert Hooke, a talented artist, architect, and amateur scientist. Hooke wrote Micrographia: Or Some Physiological Descriptions of Minute Bodies Made by Magnifying Glasses with Observations and Inquiries Thereupon, at the behest of the newly chartered Royal Society in London, for whom he was working as curator of scientific experiments. In Micrographia, he presented the first detailed observations of everyday objects made with his self-constructed light microscope....

  • Today, an individual would be hard-pressed to find any science field that does not employ methods and instruments based on the use of fine focused electron and ion beams. Well instrumented and supplemented with advanced methods and techniques, SEMs provide possibilities not only of surface imaging but quantitative measurement of object topologies, local electrophysical characteristics of semiconductor structures and performing elemental analysis.

  • At last the book is finished – and I have now been asked to put my mind to the Preface! It occurs to me that writing a Preface is a unique art form. Admittedly, after limited research into Preface-writing, I propose, like innumerable authors before me, to start with the usual whinge – yes, to paraphrase Mrs Beeton from the Preface of her famous cookbook, if we had known ‘. . . what courageous efforts were needed to be made’, I am quite sure that we would never have started this enterprise. However, it is clear that one of the...

  • With the advent of the atomic force microscope (AFM) came an extremely valuable analytical resource and technique, useful for the qualitative and quantitative surface analysis with sub-nanometer resolution. In addition, samples studied with an AFM do not require any special pretreatments that may alter or damage the sample, and permit a three dimensional investigation of the surface.

An image is formed from the interaction of the electrons transmitted through the specimen, which is then magnified and focused onto an imaging device, such as a fluorescent screen, a photographic film, or a charge-coupled device (CCD) sensor. This technique is capable of imaging at significantly high resolution than the light microscopes, owing to the small de-Broglie wavelength of electrons....
