Analyses of the spatial localization and the functions of bacteria in host plant habitats through in situ identification by immunological and molecular genetic techniques combined with high resolving microscopic tools and 3D-image analysis contributed substantially to a better understanding of the functional interplay of the microbiota in plants. Among the molecular genetic methods, 16S-rRNA genes were of central importance to reconstruct the phylogeny of newly isolated bacteria and to localize them in situ.

In this paper we present the construction of a highly sensitive FCS instrument and the measurement results from a sample of semiconductor quantum dots. We provide the analysis procedure for determining the hydrodynamic radius of the quantum dots and compare the results with the ones obtained directly from electron microscope imaging. The good agreement indicates the reliability of the FCS technique and open the way for further applications of this technique in studying nanoparticles.

The GNSs were also bioconjugated with anti-HER2 monoclonal antibody for diagnostic breast cancer cells using dark field microscope technique. These GNS NPs play a role as nanoheaters transforming light to heat. With the present of these GNS NPs at volume density of 3.6×1010 NPs.cm3 in chicken tissue samples, illuminated by 808 nm laser at the power density of 62 W.cm2 the temperature of tissue sample reachs 110˚ C after 20 minutes illumination.

The invention of scanning tunneling microscope (STM) by Binnig and his colleagues in 1982 opened up the possibility of imaging material surfaces with spatial resolution much superior to the conventional microscopy techniques. The STM is the first instrument capable of directly obtaining three-dimensional images of solid surfaces with atomic resolution. Even though STM is capable of achieving atomic resolution, it can only be used on electrical conductors. This limitation has led to the invention of atomic force microscope (AFM) by Binnig and his co-workers in 1986.

It is no surprise to see the micro-Raman Group at Lille come forth with this timely publication to document the present state of Raman microscopy. A quarter century has passed since the early attempts at Raman microsampling when the field began to merge with, and complement, other microprobe techniques. In the late 1960s to the early '70s, it was mainly the electron beam methods that opened up the microscopic domain to instrumental analysis, aside from classical light microscopy.
