Comparative analyses of chloroplast and mitochondrial genomes have shown that organelle genomes in bryophytes evolve slowly. However, in contrast to seed plants, the organellar genomes are yet poorly explored in bryophytes, especially among liverworts.

In this paper the individual markers (COI and 18S rRNA) were successfully used to identify some sponge taxa at the species level. The obtained results showed the congruence of molecular taxonomy using two independent markers. However, our study showed that a combination of the two markers provided more information and supported better for sponge identification.

Mitochondrial functional and structural impairment and generation of oxidative stress have been implicated in aging, various diseases and chemotherapies. This study analyzed azidothymidine (AZT)-caused failures in mitochondrial functions, in redox regulation and activation of the HIV-1 gene expression. We monitored intracellular concentrations of ATP and glutathione (GSH) as the indicators of energy production and redox conditions, respectively, during the time-course experiments with U937 and MOLT4 human lymphoid cells in the presence of AZT (0.05 mgÆmL)1) or H2O2 (0.01 mM) for 15–25 days.

Mitochondrial uncoupling protein 1 (UCP1) is a speci®c marker of multilocular brown adipocytes. Ectopic UCP1 in white fat of aP2-Ucp1micemitigates development of obesity byboth, increasing energy expenditure anddecreasingin situ lipogenesis. In order to further analyse consequences of respiratory uncoupling inwhite fat, the eects of the ectopic UCP1 on the morphology of adipocytes and biogenesis of mitochondria in these cells were studied.

Termination of mitochondrial (mt) H-strand transcription in mammalian cells occurs at two distinct sites on the genome. The first site of termination, referred to as mt-TERM occurs beyond the 16 S rRNA gene. However, the second and final site of termination beyond the tRNA Thr gene remains unclear. In this study we have characterized the site of termination of the polycistronic distal gene transcript beyond the D-loop region, immedi-ately upstream of the tRNA Phe gene.

The effects of physiologically relevant hypoxia on the catalytic activity of cytochromecoxidase (CytOX), mito-chondrial gene expression, and both nuclear and mito-chondrial encoded CytOX mRNA levels were investigated in murine monocyte macrophages, mouse C2C12 skeletal myocytes and rat adrenal pheochromocytoma PC12 cells. Our results suggest a coordinated down regulation of mito-chondrial genome-coded CytOX I and II and nuclear genome-codedCytOX IV andVbmRNAs during hypoxia.

Whereas the majority of disease-related mitochondrial DNA mutations exhibit significant biochemical and clinical heterogeneity, mutations within the mitochondrially encoded human cytochrome b gene (MTCYB) are almost exclusively associated with isolated complex III deficiency in muscle and a clinical presentation involving exercise intolerance.

Membrane-associated calcium-independent phospholipase A2c(iPLA2c) contains four potential in-frame methionine start sites (Mancuso, D.J. Jenkins, C.M. & Gross, R.W. (2000)J. Biol. Chem.275, 9937–9945), but the mechanisms regulating the types, amount and subcellular localization of iPLA2c in cells are incompletely understood.

Mitochondrial biogenesis is a complex and highly regulated process that requires the controlled expression of hundreds of genes encoded in two separated genomes, namely the nuclear and mitochondrial genomes. To identify regulatory proteins involved in the transcriptional control of key nuc-lear-encoded mitochondrial genes, we have performed a detailed analysis of the promoter region of theasubunit of the Drosophila melanogasterF1F0 ATP synthase complex.

The cytochromebc1 complex of the yeastSaccharomyces cerevisiae is composed of 10 different subunits that are assembledas a symmetrical dimer in the innermitochondrial membrane. Three of the subunits contain redox centers and participate in catalysis, whereas little is known about the function of the seven supernumerary subunits.

Mitochondrial fatty acid oxidation deficiencies are due to genetic defects in enzymes of fatty acidb-oxidation and transport proteins.Genetic defects have been identified in most of the genes where nearly all types of sequence vari-ations (mutation types) have beenassociatedwithdisease. In this paper, we will discuss the effects of the various types of sequence variations encountered and review current know-ledge regarding the genotype–phenotype relationship, espe-cially in patients with acyl-CoA dehydrogenase deficiencies where sufficient material exists for a meaningful discussion....

The expression of the colonic mitochondrial 3-hydroxy 3-methyl glutaryl CoA (mHMGCoA) synthase,a key con-trol site of ketogenesis from butyrate,is lower in germ-free (GF) than in conventional (CV) rats. In contrast, the activity of glutaminase is higher. The objective of this study was to investigate whether the intestinal flora can affect gene expression through short chain fatty acid (SCFA) and butyrate production. GF rats were inoculated with a con-ventional flora (Ino-CV) orwitha bacterial strainproducing butyrate (Clostridium paraputrificum,Ino-Cp) or not (Bifidobacterium breve,Ino-Bb).

Peroxiredoxins belong to a family of antioxidant proteins that neutralize reactive oxygen species. One member of this family, peroxiredoxin I (PRDX1), suppresses DNA oxidation. Peroxiredoxin V (PRDX5) has been cloned as a transcriptional corepressor, as a peroxisomal⁄mitochondrial antioxidant protein, and as an inhibitor of p53-dependent apoptosis.

The mitochondrial respiratory complex II, or succinate:ubiquinone oxido-reductase, is an integral membrane protein complex in both the tricarboxy-lic acid cycle (Krebs cycle) and aerobic respiration. The gene sequences of each complex II subunit were measured by RT-PCR. N-terminal sequen-cing work was performed to identify the mitochondrial targeting signal peptide of each subunit.

TheycaC-related gene, ycaCR, is uncharacterized, and has no assigned function to date. Here we clearly showed that the ycaC-related gene from the amphioxus Branchiostoma belcheri, BbycaCR, coded for a novel mem-ber of the isochorismatase superfamily, which is mainly localized in the mitochondrial fraction.

Peroxiredoxin V (PRDX5) is a member of the family of mammalian pro-teins that neutralize reactive oxygen species. The PRDX5gene is constitu-tively expressed at a high level in many human tissues, but functional elements of its promoter responsible for a high basal activity in the absence of oxidative stress have still not been identified.

The mechanisms underlying increased cardioprotection in younger female mice are unclear. We hypothesized that serine-threonine protein kinase (protein kinase B; Akt) triggers a metabolic gene switch (decreased fatty acids, increased glucose) in female hearts to enhance mitochondrial bio-energetic capacity, conferring protection against oxidative stress.
