Molecular characterization of endophytic bacteria
-
In the present study, 24 isolates of endophytic bacteria were obtained from different plant tissues including root, stem and fresh leaves regions of tomato plants cv. Arka Vikas. The study characterizes morphological, biochemical and molecular characters of isolated bacterial endophytes.

The aim of this study was to isolate, characterize and identify endophytic bacteria from plants growing along the stream banks in Hot Springs, South Dakota. The bacterial endophytes were isolated, identified and screened in vitrofor morphological features (Gram stain, Gram morphology, and colony morphology). Further, isolates exhibiting difference in morphological features were selected for molecular identification through partial 16S-rRNA gene sequencing.

Endophytes are the organisms that live within plant tissues and spend at least a half of their life time without causing disease under any circumstances. So, the endophytes are commonly defined as those organisms whose infections are inconspicuous and symptomless in host tissue and internal microbial colonization. Both prokaryote (bacteria) and eukaryotes (fungi) colonize the plant tissues exhibiting a little variation in their colonization mode. The former colonize intracellularly in vascular tissues and the later shows asymptomatic colonization either inter or intracellularly.

A field experiment was conducted with efficient isolates of endophytic nitrogen fixing bacteria i.e., Gluconacetobacter diazotrophicus NFGS25 and NFGM5 which were isolated from the roots of sugarcane and maize, based on the results of pot culture studies on maize. NFGS25 and NFGM5 were confirmed by the molecular characterization using 16S rDNA method. At 30 DAS(days after sowing), the growth parameters such as plant height (52.17 cm), stem girth (3.53 cm), chlorophyll content (32.5), root length (60 cm), dry matter production of shoot (3.5 g per plant) and root (0.
