The de novo design of a novel protein with a particular function remains a formidable challenge with only isolated and hard-to-repeat successes to date. Due to their many structurally conserved features, antibodies are a family of proteins amenable to predictable rational design.

Mucin, a major component of mucus, is a highly O-glycosylated, high-molecular-mass glycoprotein extensively involved in the physiology of gastrointestinal mucosa. To detect and characterize mucins derived from site-specific mucous cells, we developed a monoclonal antibody, designated PGM34, by immunizing a mouse with purified pig gastric mucin.

Prion diseases are neurodegenerative disorders which cause Creutzfeldt– Jakob disease in humans, scrapie in sheep and bovine spongiform encephalopathy in cattle. The infectious agent is a protease resistant iso-form (PrP Sc ) of a host encoded prion protein (PrP C ). PrP Sc proteins are characterized according to size and glycoform pattern.

Twenty-five years ago, Georges Köhler and César Milstein invented a means of cloning individual antibodies, thus opening up the way for tremendous advances in the fields of cell biology and clinical diagnostics (1). However, in spite of their early promise, monoclonal antibodies (MAbs) were largely unsuccessful as therapeutic reagents resulting from insufficient activation of human effector functions and immune reactions against proteins of murine origin.
