A series of novel 10-phenylsulfonyl-2-substituted-4,10 dihydrobenzo[4,5]imidazo[1,2- a]pyrimidin-4-one derivatives obtained from N-sulfonation of 2-substituted-pyrimido[1,2- a]benzimidazol-4(10H)-ones and screened for for their in vitro anti-tuberculosis activities against Mycobacterium tuberculosis H37Rv by Microplate Alamar Blue Assay (MABA) method.

A eukaryotic-type protein serine/threonine kinase, PknA, was cloned from Mycobacterium tuberculosis strain H37Ra. Sequencing of the clone indicated 100% identity with the published pknA sequence of M. tuberculosis strain H37Rv. PknA fused to maltose-binding protein was expressed in Escherichia coli; it exhibited a molecular mass of % 97 kDa. The fusion protein was purified from the soluble fraction by affinity chromatography using amylose resin.

The Rv0948c gene fromMycobacterium tuberculosisH37Rv encodes a 90 amino acid protein as the natural gene product with chorismate mutase (CM) activity. The protein, 90-MtCM, exhibits Michaelis–Menten kinetics with akcat of 5.5 ± 0.2 s )1 and a Kmof 1500 ± 100lmat 37C and pH 7.5. The 2.0 A˚ X-ray structure shows that 90-MtCM is an alla-helical homodimer (Protein Data Bank ID: 2QBV) with the topology ofEscheri-chia coliCM (EcCM), and that both protomers contribute to each catalytic site.

Pathogenicity ofMycobacterium tuberculosisis closely rela-ted to its ability to survive and replicate in the hostile envi-ronment of macrophages. For some pathogenic bacteria, secretion of ATP-utilizing enzymes into the extracellular environment aids in pathogen survival via P2Z receptor-mediated, ATP-induced death of infected macrophages. A component of these enzymes is nucleoside diphosphate kinase (Ndk). Thendkgenewas cloned fromM. tuberculosis H37Rv and expressed inEscherichia coli.

Class III adenylyl cyclases usually possess six highly conserved catalytic residues. Deviations in these canonical amino acids are observed in several putative adenylyl cyclase genes as apparent in several bacterial genomes. This suggests that a variety of catalytic mechanisms may actually exist. The geneRv0386from Mycobacterium tuberculosiscodes for an adenylyl cyclase catalytic domain fused to an AAA-ATPase and a helix-turn-helix DNA-binding domain.

The 6-kDa early secretory antigenic target (ESAT-6) and culture filtrate protein-10 (CFP-10), expressed from the region of deletion-1 (RD1) of Mycobacterium tuberculosisH37Rv, are known to play a key role in viru-lence. In this study, we explored the thermodynamic and biochemical chan-ges associated with the formation of the 1 : 1 heterodimeric complex between ESAT-6 and CFP-10.

Adenylyl cyclase Rv2212 fromMycobacterium tuberculosishas a domain composition identical to the pH-sensing isoform Rv1264, an N-terminal regulatory domain and a C-terminal catalytic domain. The maximal velo-city of Rv2212 was the highest of all 10 mycobacterial cyclases investigated to date (3.9lmol cAMPÆmg )1 Æmin )1 ), whereas ATP substrate affinity was low (SC50¼2.1 mmATP).

Mycobacterium tuberculosisH37Rv is a highly successful pathogen and its success fully relies on its ability to utilize macrophages for its replication and, more importantly, the macrophage should remain viable to host the Mycobacterium.
