  • In vitro regeneration through seeds were studied in lime (Citrus aurantifolia) by culturing lime seeds on MS media supplemented with BAP (1, 3 and 5 mg/l), Kinetin (1, 3 and 5 mg/l) and their combination with NAA (0.1, 0.5 and 1 mg/l). In reference to increased germination percentage, mean shoot length, mean number of leaves per explant and mean root length, among BAP and Kinetin concentrations, BAP 3 mg/l was found to be the best treatment. In terms of cytokinins and NAA combine effect, BAP 5 mg/l + NAA 0.5 mg/l and Kinetin 3 mg/l + NAA at 0.1 mg/l were found to be the best combinations.

  • The shoots in vitro were transferred to half-strength MS supplemented with different concentrations of IAA, IBA and NAA to investigate root formation. The best rooting occurred at 0,5 mg/L NAA (7.91 roots/shoot; root length of 4.01 cm; 98.51% root formation).   • The experiment consists of six treatments of growth regulators and control i.e. NAA 500 ppm, NAA 1000 ppm, NAA 1500 ppm, IBA 1000 ppm, IBA 1500 ppm and IBA 2000 ppm, were replicated four times in a complete randomized design (CRD).   • A study on influence of different doses of Benzyl Adenine (BA) combined with constant level of Naphthalin acetic acid (NAA) on callus induction, shoot multiplication, and rooting of Kew pineapple was conducted. Early callus response was observed in treatment concentrations of 2mg/l BA with 2mg/l NAA and 3mg/l BA with 2mg/l NAA. Treatment 2mg/l BA with 2mg/l NAA was resulted to replicate the maximum number of shoots (22.71 at 90 days) significantly among the treatments. Shoot length was most enhanced by 0.5 mg/l BA with 2 mg/l NAA (8.09 cm at 90 days) dose.

  • In vitro regeneration and in planta transformation protocols were standardized for the black gram (Vigna mungo L.) cv. VBN 3, an important grain legume. An efficient regeneration protocol was developed using 1-week-old de-embryonated cotyledon as explants.   • An effective protocol for in vitro proliferation and multiplication of Gerbera cv. Partrizia was established during 2015-17. Among the two stages initially used for culture initiation, capitulum explants cultured at immature stage responded better than the mature stage.
