Neutrophil serine proteinases
-
Inheritance of the F variant of alpha-1-antitrypsin is associated with normal circulating protein levels, but it is believed to be dysfunctional in its ability to inhibit neutrophil elastase and therefore has been implicated as a susceptibility factor for the development of emphysema.

Elafin and its precursor, trappin-2 or pre-elafin, are specific endogenous inhibitors of human neutrophil elastase and proteinase 3 but not of cathepsin G. Both inhibitors belong, together with secretory leukocyte protease inhibitor, to the chelonianin family of canonical protease inhibitors of serine proteases. AcDNAcoding either elafin or its precursor, trappin-2, was fused in framewith yeasta-factor cDNAand expressedinthePichiapastorisyeast expressionsystem.

Proteinase 3 and neutrophil elastase are serine proteinases of the polymor-phonuclear neutrophils, which are considered to have both similar localiza-tion and ligand specificity because of their high sequence similarity. However, recent studies indicate that they might have different and yet complementary physiologic roles.
