Normal phase silica gel column chromatography

Using column chromatography with normal and reverse phase of silica gel as adsorbent, a new benzofuran derivative, pumiloside (1) together wiith seven known flavonoid glycosides, afzelin (2), astragalin (3), quercitrin (4), isoquercitrin (5), kaempferol 3-O-rutinoside (6), rutin (7) and kaempferol 3-O-sophoroside (8) were isolated from the leaves of Ficus pumila.

This study aimed to investigate the antitumor-promoting effects of total ethanolic extract from Perilla leaf (PLE) in a two-stage chemical carcinogenesis protocol in mice. Quantitative analysis active compounds in PLE were performed by thin layer chromatography with normal phase of silica gel F254 and HPLC-PDA with reversed phase of C18 column at UV 300 nm. The cutaneous tumors were initiated by a single application by 7,12-dimethylbenz[a]anthracene (DMBA) on the dorsal shaved skin, then promoted by repeated applications of croton oil during 20 weeks.

The maceration was applied to the seeds of the plant to afford the crude extract which was then fractionated by liquid-liquid extraction to obtain the ethyl acetate extract. The extract was carried out by using normal phase silica gel column chromatography and thin-layer chromatography. Analysis of spectroscopic data including HR-ESI-MS, 1D and 2D-NMR and a comparison of the NMR data with that in the literature led to the structural elucidation of isolated compounds.

The lichen Roccella sinensis has not been studied chemically. This research described the isolation and elucidation of compounds isolated from the lichen Roccella sinensis collected in Binh Thuan. Phytochemistry investigation of this lichen was carried out by using normal phase silica gel column chromatography and thin-layer chromatography. Six compounds was isolated. Their structures were established by extensively spectroscopic analysis as well as comparison with NMR data in the literatures.
