In conclusion, activation by cycloheximide was the most qualified chemical for artificial activation and preimplantation development of the Bos gaurus-taurus interspecies embryo. However, H3K9me3 immunostaining can only detect the Bos gaurus pronucleus formation at the zygote stage.

Growing oocytes derived from small antral follicles (2-3 mm) have poor maturation rates and developmental competence compared to fully grown oocytes collected from large antral follicles (4-6 mm). Hence, a pre-in vitro maturation (Pre-IVM) subculture with in vitro maturation (IVM) was applied to support small oocytes growth.

The maturation and successful acquisition of developmental competence by an oocyte, the female gamete, during folliculogenesis is highly dependent on molecular interactions with somatic cells. Most of the cellular interactions identified, thus far, are modulated by growth factors, ions or metabolites. We hypothesized that this interaction is also modulated at the transcriptional level, which leads to the formation of gene regulatory networks between the oocyte and cumulus cells.

Maturation of oocytes under in vitro conditions (IVM) results in impaired developmental competence compared to oocytes matured in vivo. As oocytes are closely coupled to their cumulus complex, elucidating aberrations in cumulus metabolism in vitro is important to bridge the gap towards more physiological maturation conditions.

Cathepsin B inhibitor improves developmental competency and cryo-tolerance of in vitro ovine embryos
Cathepsin B is a lysosomal cysteine protease involved in apoptosis and oocytes which have lower developmental competence show higher expression of Cathepsin B. Furthermore, expression of Cathepsin B show a decreasing trend from oocyte toward blastocyst stage.

In the present study, effect of two different culture media on the developmental competence of bovine oocytes was studied. For the study a total of 1110 oocytes were collected using ovum pick-up (OPU) technique and the average oocyte yield was 8.34 ± 0.23 per OPU session per animal.
