Trehalose metabolism is an essential component of the stress response in yeast cells. In this work we show that the products of the principal genes involved in trehalose metabolism in Schizosaccharomyces pombe, tps1+ (coding for trehalose6-P synthase, Tps1p), ntp1+ (encoding neutral trehalase, Ntp1p) and tpp1+ (that codes for trehalose-6-P phosphatase, Tpp1p), interact in vitro with each other and with themselves to form protein complexes. Disruption of the gene tps1+ blocks the activation of the neutral trehalase induced by heat shock but not by osmotic stress.

This paper reports the results obtained using the osmotic stress method applied to the purified cathodic and anodic hemoglobins (Hbs) from the catfishHoplosternum littorale, a species that displays facultative accessorial air oxygen-ation. We demonstrate that water potential affects the oxy-gen affinity ofH. littoraleHbs in the presence of an inert solute (sucrose). Oxygen affinity increases when water activity increases, indicating that water molecules stabilize thehigh-affinitystateof theHb.

We recently cloned a novel osmotic stress transcription factor 1 (OSTF1) from gills of euryhaline tilapia (Oreochromis mossambicus) and demonstra-ted that acute hyperosmotic stress transiently increases OSTF1 mRNA and protein abundance [Fiol DF, Ku¨ltz D (2005) Proc Natl Acad Sci USA 102, 927–932].

In their aqueous habitats, fish are exposed to a wide range of osmotic con-ditions and differ in their abilities to respond adaptively to these variations in salinity. Fish species that inhabit environments characterized by signifi-cant salinity fluctuation (intertidal zone, estuaries, salt lakes, etc.) are eury-haline and able to adapt to osmotic stress.

FE65 is a neural adaptor protein known to interact with a number of pro-teins, including Alzheimer’s amyloid b-protein precursor (APP). Although several different functions have been proposed for FE65, its primary physi-ological role remains unclear. We previously showed that APP can liberate FE65 from the membrane as a result of APP phosphorylation, and that the liberated FE65 translocates into the nuclei of osmotically stressed cells.
