We describe a highly sensitive, quantitative, and inexpensive technique for targeted sequencing of transcript cohorts or genomic regions from thousands of bulk samples or single cells in parallel. Multiplexing is based on a simple method that produces extensive matrices of diverse DNA barcodes attached to invariant primer sets, which are all pre-selected and optimized in silico.

Adenosine-to-inosine (A-to-I) RNA editing is an epigenetic modification catalyzed by adenosine deaminases acting on RNA (ADARs), and is especially prevalent in the brain. We used the highly accurate microfluidics-based multiplex PCR sequencing (mmPCR-seq) technique to assess the effects of development and environmental stress on A-to-I editing at 146 pre-selected, conserved sites in the rat prefrontal cortex and amygdala. Furthermore, we asked whether changes in editing can be observed in offspring of stress-exposed rats. In parallel, we assessed changes in ADARs expression levels.

Clade 5 Clostridioides difficile diverges significantly from the other clades and is therefore, attracting increasing attention due its great heterogeneity. In this study, we used third-generation sequencing techniques to sequence the complete whole genomes of three ST11 C. difficile isolates, RT078 and another two new ribotypes (RTs), obtained from three independent hospitalized elderly patients undergoing antibiotics treatment.

In recent decade, with advances in molecular techniques, studies in AMF have turned to directly identification from soil samples by polymerase chain reaction (PCR) methods (Husband et al., 2002). In the present investigation, the diversity of AMF on medicinal plants G. sylvestre and C. longa in Vietnam was studied using PCR based method for specific AM fungal ribosomal RNA (rDNA) sequences. This paper use molecular approaches to determine and assess the distribution of AM fungi among different medicinal plant hosts.

The knowledge concerning the behavior of these Bacilli as antagonists and genetic analysis is essential for their effective use and the commercialization. The present study was focused on the analysis of the genetic diversity of rhizobacterial isolates of Bacillus using PCR based RAPD technique and selection of best biocontrol antifungal Bacillus strain with aflatoxin producing Aspergillus by antagonism on PDA medium. About 16 different strains of bacteria were isolated from healthy and infested rhizosphere of groundnut using N-agar medium.

Interrogation of site-specific CpG methylation in circulating tumor DNAs (ctDNAs) has been employed in a number of studies for early detection of breast cancer (BrCa). In many of these studies, the markers were identified based on known biology of BrCa progression, and interrogated using methyl-specific PCR (MSP), a technique involving bisulfite conversion, PCR, and qPCR.

Prevalence of Theileria equi among horses based on parasitological and universal molecular technique
Equine theileriosis, being an OIE listed disease mainly requires frequent disease screening and monitoring. The aim of the present study was to monitor the prevalence of theileriosis in horses by using both parasitological method and molecular diagnostic technique based on Equine Merozoite Antigen-5 and 6. In the present study, samples from 248 horses were collected randomly.

The objective of the study was to see the prevalence of C. perfringens as well as to detect the enterotoxigenic genes in C. perfringens strains isolated from different meat and water samples in Kashmir, India. For this 275 different meat and meat product, and water samples were analyzed by standard bacteriological technique. All the Fifty nine (59) isolates recovered from these samples were typed according to their gene by multiplex PCR assay. Out of these, 44 (74.57%) isolates were confirmed as C. perfringens type A based on the presence of alpha toxin gene (cpa).

Targeting Induced Local Lesions in Genome (TILLING) is a general reverse technique that combines chemical mutagenesis with PCR based screening to identity point mutations in regions of interest. The strategy works with a mismatch-specific endonuclease to detect induced or natural DNA polymorphisms in gene of interest. The main principles of TILLING include making EMS-mutagenized population, targeting gene of interest with PCR primers and searching rare mutants among many individuals.

Diagnosis of disease is the backbone of control and treatment in veterinary field. In addition to the antemortem and post mortem methods, currently several laboratory-based tools and technique are also being used for early diagnosis. Since few decades, polymerase chain reaction (PCR) has emerged as the most preferred molecular diagnostic technique for disease diagnosis due to its high specificity.

Tuberculosis is a major public health problem and second largest cause of death among infectious diseases. Rapid diagnosis of tuberculosis and detection of drug resistance is essential for effective disease control. Control of TB is hindered by the lack of a simple and effective diagnostic tool that can be utilised in resource limited settings. Molecular techniques using Real time PCR technology have gained momentum for rapid diagnosis and detection of drug resistance.

The aim of this study was to determine the phylogenetic relationships of Trichoderma isolates obtained from NLS Tobacco region. Utilizing the sequence analysis of internal transcribed spacer-1 (ITS-1) region of the ribosomal DNA the rDNA of Five Trichoderma isolates was amplified by polymerase chain reaction (PCR) using universal primers (ITS-1 and ITS-4). PCR products were purified and these purified products were used to amplify the ITS-4 region of the five Trichoderma isolates.

Inter Simple Sequence Repeats, the PCR based technique was used to reveal the genetic diversity among wheat and its wild relatives. DNA profiling of 27 wheat genotypes at all the existing ploidy levels (diploid, tetraploid and hexaploid) was done using ISSR primers. All the primers were polymorphic. The primers resulted in the amplification of 73 bands out of which 17 bands were unique. Unique bands resulted in case of all the primers. According to these 4 ISSR primers genetic similarity value was observed ranging from 0 to 1, the highest genetic similarity value (1.

Wheat is qualitatively a vital source of macromolecule, energy and fiber for human nutrition since decades hence, used as a staple food grain for community and as well a major source of fodder for animal feeding. Assessment of genetic diversity using molecular markers is used for characterization of different genotypes with reliable and authentic results. RAPD is a PCR based molecular technique for identification of genetic variability in similar genotypes. It usually preferred for the initiation of this kind of work as this technique is simple, versatile and relatively inexpensive.
