In ICU patients, glomerular filtration is often impaired, but also supraphysiological values are observed (“augmented renal clearance”, >130 mL/min/1.73 m2 ). Renally eliminated drugs (e.g. many antibiotics) must be adjusted accordingly, which requires a quantitative measure of renal function throughout all the range of clinically encountered values.

Pharmacokinetic estimates for intravenous paracetamol in individual adult cohorts are different to a certain extent, and understanding the covariates of these differences may guide dose individualization. In order to assess covariate effects of intravenous paracetamol disposition in adults, pharmacokinetic data on discrete studies were pooled.

Many medical imaging techniques utilize fitting approaches for quantitative parameter estimation and analysis. Common examples are pharmacokinetic modeling in dynamic contrast-enhanced (DCE) magnetic resonance imaging (MRI)/computed tomography (CT), apparent diffusion coefficient calculations and intravoxel incoherent motion modeling in diffusion-weighted MRI and Z-spectra analysis in chemical exchange saturation transfer MRI.

Present study was conducted for pharmacokinetic interaction of long acting moxofloxacin (LA moxifloxacin) with meloxicam at the dose rate of 7.5 mg.kg-1 and 0.5 mg.kg-1 respectively following intramuscular (IM) administration. Peak plasma concentrations of drug were estimated by using HPLC florescence detector

Highly sensitive and selective liquid chromatography/tandem mass spectrometry (LC–MS/MS) method was developed and validated for the simultaneous estimation of the recently approved oral hypoglycemic mixture; metformin (MET) and canagliflozin (CFZ) in human plasma using propranolol HCl (PPL) and tadalafil (TDF) as internal standards (IS), respectively.

AR-42, a new orally bioavailable, potent, hydroxamate-tethered phenylbutyrate class I/IIB histone deacetylase inhibitor currently is under evaluation in phase 1 and 2 clinical trials and has demonstrated activity in both hematologic and solid tumor malignancies. This report focuses on the preclinical characterization of the pharmacokinetics of AR-42 in mice and rats.

The physiologically based pharmacokinetic (PBPK) model for liver transporter substrates has been established previously and used for predicting drug–drug interactions (DDI) and for clinical practice guidance. So far, nearly all the published PBPK models for liver transporter substrates have one or more hepatic clearance processes (i.e., active uptake, passive diffusion, metabolism, and biliary excretion) estimated by fitting observed systemic data.

Gender-based pharmacokinetics and/or pharmacodynamics differences can result in differences in treatment which can accordingly affect the drug safety and/or efficacy. A new validated bio-analytical LC-MS/ MS method was developed for the estimation of ezogabine, a third-generation antiepileptic drug, in human plasma using oxcarbazepine as an internal standard (IS) and to study the gender effect on the pharmacokinetic parameters in Egyptian human subjects.

This study aimed to estimate PKparameters of imipenem and those potential covariates. Methods: Burn patients with body surface area injured >20% andimipenem indicationwere recruited. Two set of plasma samples (30 min post-dose and 1-2 hours before next dose)were obtainedat imipeneminitiation and before the end of imipenem use. ARC was defined if 8h-urinary creatinine clearance (8hClcr) was above 130 mL.min1173 m2.

The subspecialty of population pharmacokinetics was introduced into clinical pharmacology / pharmacy in the late 1970s as a method for analyzing observational data collected during patient drug therapy in order to estimate patient-based pharmacokinetic parameters. It later became the basis for dosage individualization and rational pharmacotherapy. The population pharmacokinetics method (i.e., the population approach) was later extended to the characterization of the relationship between pharmacokinetics and pharmacodynamics, and into the discipline of pharmacometrics.

Despite massive efforts in drug discovery fueled by combinatory chemistry, recombinant DNA technology, and highthroughput screening, surprisingly few molecules make it through the drug development process. While the reasons are debated, it is certain that many new chemical entities (NCEs) suffer from recurring problems that hinder development— low water solubility, instability, or inadequate pharmacokinetics. An estimated 43% of NCEs are poorly water-soluble.
