The study aimed to explore the molecular mechanisms of miR-124-3p which influence IR leading to T2DM establishment. HepG2 cells were cultured in vitro, and palmitic acid (PA) was used to construct the IR cell model. In the IR model, transfection of miR-124-3p or phosphoglucomutase 1 (PGM1) linked plasmids were transfected into HepG2 cells. RT-qPCR was used to determine the miR-124-3p and PGM1 expressions in the cells.

PIKfyve is an evolutionarily conserved lipid and protein kinase enzyme that has pleiotropic cellular functions. The aim of the present study was to investigate the effects of phosphatidylinositol-3-phosphate 5-kinase (PIKfyve) inhibitor, YM201636, on nonsmall cell lung cancer (NSCLC) cells growth, tumorigenicity, and claudin (CLDN) expressions. Three NSCLC cell lines (Calu-1, H1299 and HCC827) were used to compare the effects of YM201636. Cytotoxic effects of YM201636 were analysed using XTT assay.

Doxorubicin (Dox) is widely used to treat progressed bladder cancer after transurethral resection. The use of Dox-chemotherapy has been limited due to induced drug resistance and cumulative cardiotoxic effects. N-benzyladriamycin-14-valerate (AD198), a novel derivative of Dox, has a potential to become a more effective treatment than Dox by overcoming drug resistance and cardio-toxicity as shown in the rodent model of lymphoma in vivo.

The protein-modifying agent arsenite stimulates glucose uptake in 3T3-L1 adipocytes. In the current study we have analysed the signalling pathways that contribute to this response. By subcellular fractionation we observed that arsenite, like insulin, induces translocation of the GLUT1 and GLUT4 glucose transporters from the low-density membrane fraction to the plasma membrane. We used two inhibitors of the signaling enzyme phosphatidylinositol 3-kin-ase (PtdIns3K), wortmannin and LY294002, to evaluate the potential involvement of PtdIns3K in the activation of the MAP kinases (MAPK), Erk1 and Erk2. In dose–response studies carried out on six different cell lines and a primary cell culture, we analyzed the ability of the inhibitors to block phosphorylation of protein kinase B⁄akt (PKB⁄akt) at Ser473 as a measure of PtdIns3K activity, or the phosphorylation of Erk1⁄2 at activa-ting Thr⁄Tyr sites as a measure of the extent of activation of MAPK⁄Erk kinase (MEK⁄Erk).
