  • Fusarium oxysporum f. sp. cumini is a serious disease of cumin (Cuminum cyminum L.). Ten isolates of F. oxysporum f. sp. cumini were isolated from major cumin growing districts of Gujarat state to compare molecular methods over traditional for differentiation among the isolates of pathogen. The average 98.70 % of polymorphism was recorded for all the 11 ISSR primers. The polymorphic information content (PIC) value ranged from 0.571 to 0.831 with an average of 0.739 per primer. The IISR primers grouped majority of the isolates based on their geographical location.

  • A set of 45 polymorphic microsatellite markers, representing markers for almost each chromosome, was used for the assessment of genetic diversity in 35 genotypes of bread wheat. A total of 127 alleles were detected which ranged from 2 to 6 with an average allele number of 2.82 per locus. The PIC value of the 45 SSRs ranged from 0.08 to 0.50 with an average PIC of 0.30. Resolving power and Marker index of all 45 markers varied from 0.17 to 2.82 and 0.16 to 1.52, respectively.

Twenty (20) primers were screened, out of which 9 primers gave polymorphic bands which were used for further analysis.   • Molecular diversity analysis on 8 hybrids of tomato generated 27 polymorphic markers. The major allele frequency for the marker SSR1- 23.2 and SSR5-13.1 was least. The polymorphic information content value (PIC) ranged from 0.00 (SSR1-23.2) to 0.86 (SSR1-62.2) with an average PIC value of 0.46 per marker indicating more diversity at DNA level. Alleles ranged from 1-8 per locus with an average of 3.33 alleles per locus. Cluster analysis resulted into 3 main clusters and 4 subclusters. The diverse genotypes can be used for hybridization programmes.

  • Bread wheat is most cultivated cereal used as food over 95% of the population. Twenty wheat genotypes were assayed to study the genetic diversity using molecular markers. The seventy-five alleles were identified with a mean of 2.34 alleles per locus using 32 SSR markers. Majority of SSR markers showed a high level of polymorphism. PIC values ranged from 0.05 (WMS-169) to 0.75 (CWM-107), with an average of 0.38 per primer. The RP value of primer ranges from 0.92 (WMC-177) to 1.94 (WMS-169) with an average value of 1.51, which explains the ability of primers to resolve the studied germplasm.

  • Genetic diversity was performed among F2 plants of the cross RH 30×CS 52 in Indian mustard (Brassica juncea) (CS 52 is salinity tolerant and RH 30 is salinity susceptible) using SSR markers. Out of 358 SSR markers, 42 were found polymorphic and 154 were monomorphic. A total of 225 alleles, ranging from 2 to 4 were amplified. The PIC (Polymorphic Information Content) value ranged from 0.427-0.730 of Jaccard’s similarity coefficients was generated between these F2 populations. F2 population was grouped in 2 major clusters and 37 sub-clusters at a similarity coefficient of 0.58.

  • The genetic diversity and population structure in 51 chickpea accessions were studied using 30 chickpea specific SSR markers. Twenty eight SSR markers exhibited polymorphism producing a total of 217 alleles. The average number of alleles per locus was 7.75. The average PIC value was 0.75 and ranged from a minimum of 0.53 (TA1) to a maximum of 0.85 (TA64). The markers TA5, TA14, TA18, TA21, TA64, TA71, TA106, TR20, TR26, TR58, TS43 were considered to be highly informative (PIC ≥ 0.8), in evaluating allelic variation present in the chickpea accessions.

  • Genetic diversity assessment is necessary to help tackle the threats of environmental fluctuations and for the effective exploitation of genetic resources in breeding program. Recent advancement in the field of molecular markers has made the genetic characterization of genotypes rapid, reliable and reproducible.   • An experiment was carried out to study the divergence among twenty four rice genotypes (14 indica and 10 japonica lines) for 15 agro-morphological traits using D2 statistics and at molecular level using 114SSR markers. The cluster analysis using Mahalonobis’ D2 statistics classified the genotypes into five clusters. The cluster IV was the largest with seven indica and three tropical japonica types.   • Information on allelic differentiation is very important to elucidate the nature and extent of divergence among Brassica juncea genotypes. In present study 20 Brassica juncea L. genotypes generated 79 polymorphic alleles from 15 SSR primer pairs with an average of 5.26 alleles per locus.   • Genetic diversity using Random Amplified Polymorphic DNA (RAPD) markers was assessed in forty eight Indian potato germplasm.   • Seed dormancy is an important trait affecting grain yield and quality in cereal crops. It is controlled by its own complicated genetic and environmental factors. The polymorphism pattern study of 24 rice genotypes for RM primers used in the present molecular study were clearly classified the rice genotypes into dormant (Cluster I, Cluster IIA) and nondormant (Cluster II B, Cluster III).   • A total of 200 SSR markers from different Brassica species were used in this study. Out of 200 SSR markers analyzed for polymorphism in two parental Brassica juncea genotypes (RB 50, drought tolerant and Kranti, drought susceptible), 51 were polymorphic. The polymorphic markers were used to screen F2 population. A total of 108 alleles were identified in the RB 50 and Kranti and the parental B. juncea genotypes. The PIC (polymorphic information content) values for various primers ranged from 0.340-0.505 with an average of 0.406.

  • The PCR products obtained from SSR analysis were separated on 3 percent high resolution agarose gel and the amplification patterns were observed. Eleven SSR primers which showed maximum polymorphism were selected for fingerprinting. The amplification pattern obtained with these primers were scored and depicted to develop fingerprint for each variety. Most of the amplicons were found to be shared among the genotypes. However, the pattern of sharing was different and good enough to separate out most of the varieties.
