The protein-modifying agent arsenite stimulates glucose uptake in 3T3-L1 adipocytes. In the current study we have analysed the signalling pathways that contribute to this response. By subcellular fractionation we observed that arsenite, like insulin, induces translocation of the GLUT1 and GLUT4 glucose transporters from the low-density membrane fraction to the plasma membrane. Arsenite did not activate early steps of the insulin receptor (IR)-signalling pathway and the response was insensitive to inhibition of phosphatidylinositol-3¢-kinase (PI-3¢)kinaseby wortman-nin. ...

In the yeast Saccharomyces cerevisiae, plasma membrane H + -ATPase is activated by d-glucose. We found that in the absence of glucose, this enzyme forms a complex with acetylated tubulin. Acetylated tubulin usu-ally displays hydrophilic properties, but behaves as a hydrophobic com-pound when complexed with H + -ATPase, and therefore partitions into a detergent phase.

PI3K is a heterodimeric lipid kinase that catalyses the conversion of phosphatidylinositol bisphosphate (PIP2) to phosphatidylinositol trisphosphate (PIP3), which acts as a plasma membrane docking site for proteins that contain a pleckstrin homology (PH) domain. These include the serine/threonine kinases Akt and PDK1 that are key downstream effectors of PI3K action (Fig. 80-2). Diagnostic Tests The choice of diagnostic tests should be guided by the history and the physical examination. Measurements of serum electrolytes, glucose, and the hematocrit are usually indicated. Withdrawal of drugs, if abrupt, can cause disease, e.g. benzodiazepines, antiepilepsy drugs. • Drugs can conceal disease, e.g. adrenal steroid. • Drugs can interact causing positive adverse effect, or negative adverse effect, i.e. therapeutic failure. • Drugs can give diagnostic clues, e.g. ampicillin and amoxicillin causing rash in infectious mononucleosis — a diagnostic adverse effect, not a diagnostic test.
