PME activity

In this study, plums were coated with lac based (2:3), SemperfreshTM (1:3), vegetable wax (1:5) and water dip (control) just after harvest on field as on-farm treatment and after transportation to the lab as off-farm treatment. After coating, the fruits were packed in punnets and stored under cold storage conditions (2 ± 1°C) at 90–92% relative humidity.

Three sweet pepper varieties viz. Swarna, California wonder and Oroballe were treated with derivatives of salicylic acid viz. Acetyl salicylic acid (ASA), 5 Sulpho salicylic acid (5 SSA) and Gentisic acid (GA) at 0.5 mM and 1 mM concentrations under ambient condition (20±5°C) and analyzed for their change in physical and biochemical profile at 3 days interval for 9 days. Fruits treated with 1 mM acetyl salicylic acid and gentisic acid significantly delayed the senescence process by delaying the changes of weight, and firmness.

A simple and effective procedure was developed to extract pectin methylesterase (PME) from Valencia orange peels. Orange peels contain 25-34 μmol of COOH min−1 g−1 of peel PME activity. The enzyme was ionically bound to cell walls and could not be extracted with water.
