The present investigations was carried out for stone characterization, analyze the physicochemical properties of media and to identify the best suitable media for germination using different polyembryonic rootstocks of mango during 2012-13 at Regional Horticultural Research and Extension Centre, Bengaluru. The studies revealed that, the significant variation was found in stone attributes (weight, length, breadth, circumference and volume).

The protocol for high frequency induction of callus from nucellus tissue was re initiation attained in polyembryonic mango cultivars Vellaikolumban and Olour. Among different sterilization treatments followed, dipping fruits in 70 % ethanol +followed by treating them in Cetrimide 1000 ppm - 15 minutes further treating with NaOCl - 1 % +Tween 20 (one drop per 100 ml) for 15 minutes and Flame sterilisation under laminar air flow found to be the best treatment.

Study on the seedling progenies of three polyembryonic varieties was carried out to differentiate zygotic and nucellar seedlings through molecular characterization. The fingerprinting showed variation across the varieties of selected seedling progenies. The variety Peach exhibited 100% zygotic seedlings among the varieties screened. The variety Nekkare was found to be 36.84% zygotic and minimum number of zygotic seedlings (10.52 %) was observed in Bappakkai.
