PPR virus

A total of 119 different clinical samples (nasal swab, conjunctival swabs, oral swabs and tissue samples) from sheep and goats were collected from the area under study for detection of PPR antigen by Sandwich-ELISA. Out of 119 clinical samples, 37 samples were found positive in small ruminants by S-ELISA, giving an overall incidence rate of 31.09 % (37/119).

A prevalence study of PPR among small ruminants was conducted in five districts (Udaipur, Dungarpur, Rajsamand, Chittorgarh and Banswara) of southern Rajasthan including both vaccinated and unvaccinated flocks of sheep and goats. The overall seroprevalence of PPR virus antibodies in vaccinated small ruminants is 42% with highest prevalence in Chittorgarh (54.95%) and lowest in Banswara (12%). In unvaccinated group, the overall seroprevalence is 42.18% with highest prevalence in Banswara (66.07%) and lowest prevalence in Udaipur district (19.04%).

Region wise monitoring of the PPR virus by active surveillance is essential as it aids in strategic planning of measures of disease control and prevention. The North Eastern Karnataka with sizable sheep and goat population experiences the outbreaks of PPR but the attempts to isolate and characterize the prevailing PPR virus is limited. Hence, the present research work was carried out with the objective of isolation and characterization of PPR virus isolates circulating in North Eastern Karnataka by microscopic (TEM) and molecular (RT-PCR) methods.

Peste-des-petits ruminants (PPR) is an economically devastating viral disease of sheep and goats. In the present study, we have evaluated the reactivity profile of the recombinant non-structural protein C of PPR virus with vaccinated and convalescent goat sera in an indirect ELISA. The antibody against this protein is getting produced in both vaccinated as well as in convalescent animals.

Peste-des-petits-ruminants (PPR) is a transboundary viral disease of small ruminants. In the present study, the full length C gene was amplified from PPR vaccine virus (Sungri/96) and expressed in a prokaryotic expression system. Specificity of the recombinant C protein was confirmed through immune-blot using anti-His conjugate and hyperimmune sera from goats. The immunogenicity of the protein was assessed using monospecific sera raised in goats. The recombinant C protein produced in this study will be useful for the development of DIVA enabled vaccine and diagnostics.

Peste-des-petits Ruminants (PPR) is a disease of small ruminants especially goat and its control and eradication till 2030 wants an extensive research to develop a potent vaccine. The role of surface HN protein in the attachment of the virus to cellular receptors makes it an appropriate target to develop a theranostics against the virus. In this study, cytotoxic T cells epitopes that will bind to MHC class I alleles were predicted out using bioinformatic tools.

The cytotoxicity and virus titration of chitosan nanoparticles encapsulated live PPR virus vaccine (PPRV-CS-NPs) was analyzed in this study. Live attenuated PPR virus vaccine (AR-87) was encapsulated in chitosan nanoparticles using ionic gelation technique. The PPRV-CS-NPs were tested for cytotoxicity study in Vero cells with different concentrations (1-40g).The cell viability was more than 80 per cent with all the concentration of the PPRV-CS-NPs tested. This indicates that the PPRV-CS-NPs are not toxic even in concentration higher than the required vaccine dose.
