Primary drug resistance

5-Fluorouracil (5FU) is a primary chemotherapeutic agent used to treat oral squamous cell carcinoma (OSCC). However, the development of drug resistance has significantly limited its clinical application. Therefore, there is an urgent need to determine the mechanisms underlying drug resistance and identify effective targets.

Paclitaxel, a tubulin-binding agent, is a Food and Drug Administration-approved first-line drug for the treatment of non-small cell lung cancer (NSCLC), for both squamous and non-squamous cell lung carcinoma, with paclitaxel/carboplatin+bevacizumab a common chemotherapy regimen for stage IV non-squamous NSCLC; however, primary or acquired resistance to paclitaxel is gradually increasing, leading to treatment failure.

A major advance in antimalarial drug discovery has been the shift towards cell-based phenotypic screening, with notable progress in the screening of compounds against the asexual blood stage, liver stage, and gametocytes. A primary method for drug target deconvolution in Plasmodium falciparum is in vitro evolution of compound-resistant parasites followed by whole-genome scans.

Pulmonary pleomorphic carcinoma is a very rare histological type of primary lung cancer, and usually provides aggressive clinical courses. A 65-year-old Japanese woman was diagnosed by transbronchial biopsy of the primary tumor as c-stage IV (cT4N3M1b) of adenocarcinoma harboring L858R point mutation in the exon 21 of epidermal growth factor receptor (EGFR).

Targeted therapies for Primary liver cancer (HCC) is limited to the multi-kinase inhibitors, and not fully effective due to the resistance to these agents because of the heterogeneous molecular nature of HCC developed during chronic liver disease stages and cirrhosis.

The microbiology and the susceptibility patterns of infected total knee arthroplasties (TKAs) vary depending on demographic, local antimicrobial stewardship, and surgical factors. We wanted to compare the recent microbial profile and antimicrobial resistance pattern in revisions due to infections after primary TKAs in Sweden and Lithuania.

Recently reported data have suggested that only a small subset of cancer cells possess the capability to initiate malignancies. These observations were based on investigation of cells within the primary tumors displaying a distinct surface marker pattern. CD133 marker is a putative hematopoietic and neuronal stem cell marker, which is also considered to be a tumorigenic marker in brain, prostate and liver. Recent studies have shown that a small population of CD133-positive cells, indeed, exists in human hepatocellular carcinoma (HCC) cell lines and primary HCC tissues.

We investigated the role of the HIPK2–p53 signaling pathway in tumorigenesis and resistance to the drug Verbascoside (VB) in colorectal cancer (CRC), using in vivo and in vitro experiments. Methods: Primary human CRC samples and normal intestinal tissues from patients were analyzed for HIPK2 expression by immunohistochemistry (IHC) and its expression was correlated against patients’ clinicopathological characteristics.

The management of ovarian cancer remains a challenge. Because of the lack of early symptoms, it is often diagnosed at a late stage when it is likely to have metastasized beyond ovaries. Currently, platinum based chemotherapy is the primary treatment for the disease. However acquired drug resistance remains an on-going problem.

Cisplatin (DDP)-based chemotherapy is the common first-line therapy for lung cancer. However, their efficacy is often limited by primary drug resistance and/or acquired drug resistance. The aim of this study was to investigate the function of miRNA-146a (miR-146a) in DDP-resistant non-small cell lung cancer (NSCLC), as well as the underlying mechanisms.

Despite the remarkable advances in the early diagnosis and treatment, overall 5-year survival rate of patients with pancreatic cancer is less than 10%. Gemcitabine (GEM), a cytidine nucleoside analogue and ribonucleotide reductase inhibitor, is a primary option for patients with advanced pancreatic cancer; however, its clinical efficacy is extremely limited.

The prevalence of multi drug resistant gram negative bacteria has increased continuously over the past few years, of particular concern are strains producing AmpC β-lactamases. The irrational uses of antibiotics are the primary cause of the increase in drug resistant bacteria. Several studies have reported that AmpC producers are isolated mainly from patients with prolonged hospitalization, but certain recent studies have reported that AmpC producers was also isolated from long term care facilities and outpatient clinics, which indicate their presence in the community.

Urinary tract infection (UTI) is considered as the most common bacterial infectious disease seen among the pediatric patients. Pediatric urinary tract infections are associated with high morbidity and long term complications like renal scaring, hypertension and chronic renal failure The epidemiology of UTI during childhood varies by age, gender and other factors (Bickerton and Ducket, 1985). Most commonly bacteria causes of UTI in children are members of Enterobacteriaceae, particularly uropathogenic strains of E. coli and Klebsiella spp.

Staphylococcus haemolyticus is part of the human normal skin microflora and is also found in nonhuman primates. This organism has been documented as a cause of primary and nosocomial bacteremia in clinical health care settings. Though CONS is ruled out as a contaminant, the emergence of multidrug-resistant isolates to glycopeptides, methicillin, fluoroquinolones, macrolides, lincosamides, streptogramin B and linezolid results in treatment failure in primary bacteremia.

Non-fermenting gram-negative bacilli (NFGNB) are an emerging problem in Blood stream infections. A major concern is multi-drug resistance which severely limits treatment options. Earlier it was believed to be non pathogenic, but recently they are more frequently isolated as primary pathogen. Usually they cause hospital acquired infection (HAI). A prospective study was conducted to isolate the NFGNB from blood samples, to identify the risk factors leading to blood stream infections and to determine the antibiotic susceptibility pattern of them.
