Produce cloned embryos

This study aimed to establish culture medium changing system to improve the efficiency of bovine somatic cell nuclear transfer protocol. Our results showed that using the culture medium of Takahashi and First 1992 produced the cloned bovine blastocyst at the highest rate (9.4%, P < 0.05) among different modified synthetic oviduct fluid (mSOF) media.

Animal cloning is the process of producing an entire organism that theoretically has the same genome as its donor cell line. To validate the success of SCNT, one must verify that the genetic source of the cloned organism is identical to the donor cell. However, none of the studies have performed to identify the genetic characteristics in the stage of preimplantation development of cloned embryo.

In this study, oocytes were recovered from 200 numbers of goats’ ovaries from different slaughterhouses for subsequent use in-vitro maturation, enucleation, reconstruction and embryo culture to produce cloned embryos. The collected oocytes were classified into three categories (Grade A, B and C) based on their appearance of homogenous granular ooplasm and layers of cumulus cells surrounding oocytes under a stereozoom microscope. The average oocyte recovery, In-vitro maturation and the enucleation rates were found to be 3.36 per ovary, 60.37 per cent and 75.52 per cent respectively.
