Lymphoma represents a heterogeneous group of cancers affecting the lymphatic system, encompassing nonHodgkin lymphoma (NHL) and Hodgkin lymphoma (HL). The majority of NHL cases originate from B cells. HL is characterized by the presence of rare Hodgkin and Reed-Sternberg cells. Within the context of the inflammatory response, two SH2 domain-containing protein tyrosine phosphatases (SHPs), namely SHP-1 and SHP-2, serve as negative regulators.

Immature mammalian oocytes are held arrested at prophase I of meiosis by an inhibitory phosphorylation of cyclin-dependent kinase 1 (CDK1). Release from this meiotic arrest and germinal vesicle breakdown is dependent on dephosphorylation of CDK1 by the protein, cell cycle division 25B (CDC25B).

The high risk Human Papillomavirus (HPV) E6 oncoproteins play an essential role in the development of cervical malignancy. Important cellular targets of E6 include p53 and the PDZ domain containing substrates such as hScrib and Dlg. We recently showed that hScrib activity was mediated in part through recruitment of protein phosphatase 1γ (PP1γ).

Tumor-suppressor genes are inactivated by methylation in several cancers including acute myeloid leukemia (AML). Src homology-2 (SH2)-containing protein-tyrosine phosphatase 1 (SHP-1) is a negative regulator of the JAK/STAT pathway.

Modulation of protein activity by phosphorylation through kinases and subsequent dephosphorylation by phosphatases is one of the most prominent cellular control mechanisms. Thus, identification of kinase substrates is pivotal for the understanding of many – if not all – molecular biological processes.

The genetic markers in inflammatory responses during mastitis afford a reasonable way for improving milk production in the Egyptian buffalo breed. Among them is the interleukin 8 Receptor Gene (IL8RB) (CXCR2); a chemokine receptor gene augments the neutrophil migration during infection. To understand its role better during mastitis in Egyptian buffalos, twenty-five dairy animals representing the normal, sub-clinically, clinically and chronically affected buffalos were randomly selected from different districts.

Inhibitors of protein tyrosine phosphatase 1B (PTP1B) are promising agents for the treatment of type 2 diabetes and obesity. From the alcoholic extract of the aerial parts of Selaginella tamariscina, four compounds including selaginellin (1), amentoflavone (2), βsitosterol (3), and scoparone (4) have been isolated and structurally elucidated by using combined chromatographic and spectroscopic experiments. Among the isolates, selaginellin (1) and amentoflavone (2), exhibited potential PTP1B inhibitory activities with IC50 values of 15.9 ± 0.6 and 7.4 ± 0.5 µM, respectively.

SHP-1, a protein-tyrosine phosphatase with two srchomology 2 domains, is expressed predominantly in hematopoietic and epithelial cells and has been implicated in numerous signaling pathways as a negative regulator. Two promoters direct the expression of human and murine SHP-1, and two types of transcripts (I) and (II) SHP-1, are initiated from each of these promoters. The cDNA sequences of (I)SHP-1 and (II)SHP-1 are identical except in the 5¢ untranslated region and in the first few coding nucleotides.

The association of tubulin carboxypeptidase with micro-tubules may be involved in the determination of the tyrosi-nation state of the microtubules, i.e. their proportion of tyrosinated vs. nontyrosinated tubulin. We investigated the role of protein phosphatases in the association of carb-oxypeptidase with microtubules in COS cells. Okadaic acid and other PP1/PP2A inhibitors, when added to culture medium before isolation of the cytoskeletal fraction, pro-duced near depletion of the carboxypeptidase activity asso-ciated with microtubules. ...

Stimulation of glycogen-targeted protein phosphatase 1 (PP1) activity by insulin contributes to the dephosphorylation and activation of hepatic gly-cogen synthase (GS) leading to an increase in glycogen synthesis. The gly-cogen-targeting subunits of PP1, GLand R5⁄PTG, are downregulated in the livers of diabetic rodents and restored by insulin treatment. We show here that the mammalian genePPP1R3Eencodes a novel glycogen-target-ing subunit of PP1 that is expressed in rodent liver.

There is mounting evidence that the vanilloid (capsaicin) receptor; transient receptor potential channel, vanilloid subfamily member 1 (TRPV1), is subjected to multiple interacting levels of control. The first level is by reversible phosphorylation catalyzed by intrinsic kinases (e.g. protein kinase A and C) and phosphatases (e.g. calcineurin), which plays a pivotal role in receptor sensitization vs. tachyphy-laxis.

PTP-S2⁄TC45 is a nuclear protein tyrosine phosphatase that activates p53 and induces caspase 1-dependent apoptosis. We analyzed the role of ICE protease-activating factor (Ipaf), an activator of caspase 1 in p53-depend-ent apoptosis. We also determined the sequence of events that lead to apoptosis upon caspase 1 activation by Ipaf.

Src homology 2 (SH2) domain-containing protein tyrosine phosphatase-1 (SHP-1) is a critical inhibitory regulator in T cell-receptor (TCR) signaling. However, the exact molecular mechanism underlying this is poorly defined, largely because the physiological substrates for SHP-1 in T cells remain elusive.

The RCAN1 protein (previously called calcipressin 1 or MCIP1) binds to calcineurin, a serine⁄threonine phosphatase (PP2B), and inhibits its activ-ity. Here we demonstrate that regulated overexpression of an RCAN1 transgene (this gene was previously called DSCR1orAdapt78) also stimu-lates expression of the GSK-3bkinase, which can antagonize the action of calcineurin.
