RAPD molecular marker based genetic

Soybean is an important legume and oilseed crop with high protein (40%) and oil (20%). RAPD markers were used to access the genetic diversity among twenty four soybean genotypes. A total of Twenty primers were used out of which 18 got amplified which produced 164 bands and all were found polymorphic i.e. 100% polymorphism. The total number of amplified bands varied between 2 (OPF-19) and 16 (OPA-01) with an average of 9.1 bands per primer.

Two molecular techniques, RAPD and ISSR were used to study the genetic relatedness in 37 genotypes of chrysanthemum. With 27 RAPDs and 10 ISSRs, a total of 271 and 107 polymorphic bands were generated accounting to 97.4% and 93.86% polymorphism respectively. Both kinds of markers could able to distinguish all the genotypes. Based on RAPD, ISSR and combination of RAPD and ISSR markers, dendrograms were constructed according to Jaccard’s Coefficient of similarity.

Oriental pickling melon (Cucumis melo var. conomon) is one of the melon group vegetables of the Cucurbitaceae, with high potential as commercial short duration vegetable in South India. Twenty five RAPD primers were used which generated a total of 182 bands, out of which 107 bands were polymorphic (58%) revealing a substantial amount of genetic diversity within this species. Similarity values among all the accessions ranged from 0.948 to 0.619.

Twenty five isolates of Trichoderma harzianum and Trichoderma viride were obtained from rhizosphere soil of turmeric and ginger collected from Belagavi and Bagalkot districts of Karnataka and eight potential isolates based on antagonistic activity were studied using RAPD PCR. The genetic relatedness among two isolates of Trichoderma harzianum and six isolates of Trichoderma viride were analysed with five random primers. RAPD profiles showed genetic diversity among the isolates with the formation of two major clusters.

Genetic variability among the 24 isolates of S. rolfsii was studied by using molecular markers like ITS-PCR and RAPD primers. Amplification of ITS region of rDNA with specific ITS1 and ITS4 universal primers produced approximately 650 to 700 bp in all the isolates of the fungus confirmed that all the isolates obtained are Sclerotium rolfsii and were sequenced. Identity of the isolates was confirmed with sequences of NCBI data base of S. rolfsii.

Random Amplified Polymorphic DNA (RAPD) Markers were use to determined the genetic variability of eight divers cowpea (Vigna unguiculata L. walp) genotypes from different eco-geographical regions of India. Total primers 40 out of there 35 could be generated DNA fragment were 1479 but were as 1159 showed polymorphic pattern with 78.34 per cent. The polymorphism was scored and used in band sharing analysis to identify genetic relationship between the genotypes.

Genetic diversity using Random Amplified Polymorphic DNA (RAPD) markers was assessed in forty eight Indian potato germplasm. Twenty RAPD markers amplified a total of ninety-one different loci that exhibited 82.30 per cent polymorphism among 48 potato germplasm. The PIC value ranged from 0.27 to 0.55. All the loci amplified by the primer which were found to be polymorphic varied in size from 1500bp. The Jaccard’s similarity coefficient was found to vary from 0.22 to 0.87. The desi cotton hybrids viz., G.Cot.DH-7 (Sujay x G 27) and G.Cot.DH-9 (4011 x 824) and their parents were characterized using morphological and molecular markers. The desi cotton hybrids and their parents showed variations for morphological characters viz., foliage, leaf shape, leaf hairiness, growth habit, pollen colour, bolls shape and stem hairiness. Based on the distinct morphological characters one can identify true hybrid. A set of 25 RAPD and 19 ISSR were used for molecular characterization of desi hybrids.
