The rearrangements in the 22q11.2 chromosomal region, responsible for the 22q11.2 deletion and microduplication syndromes, are frequently associated with congenital heart disease (CHD). The present work aimed to identify the genetic basis of CHD in 87 patients from the São Miguel Island, Azores, through the detection of copy number variants (CNVs) in the 22q11.2 region.

Chromatin loops form a basic unit of interphase nuclear organization, with chromatin loop anchor points providing contacts between regulatory regions and promoters. However, the mutational landscape at these anchor points remains under-studied. Here, we describe the unusual patterns of somatic mutations and germline variation associated with loop anchor points and explore the underlying features influencing these patterns.

Mitochondrial genomes of flowering plants (angiosperms) are highly dynamic in genome structure. The mitogenome of the earliest angiosperm Amborella is remarkable in carrying rampant foreign DNAs, in contrast to Liriodendron, the other only known early angiosperm mitogenome that is described as ‘fossilized’. The distinctive features observed in the two early flowering plant mitogenomes add to the current confusions of what early flowering plants look like.

Sorghum shoot fly [Atherigona soccata (Rondani)] is one of the most important pests of grain sorghum. It is important to understand the nature of gene action to breed for resistance to the target insect pest. In the present study, a population of 259 sorghum recombinant inbred lines (RILs) derived from cross 296B (shoot fly susceptible) x IS 18551 (shoot fly resistant) were evaluated in shoot fly screening nursery to study the inheritance of shoot fly resistance. Variances due to RIL genotypes (G), screening environment (E) and G x E interactions were significant for the traits studied.

Urease (urea amidohydrolase, EC 3.5.1.5) is a nickel-containing enzyme produced by plants, fungi, and bacteria that catalyzes the hydrolysis of urea into ammonia and carbamate. Urease is of historical importance in Biochemistry as it was the first enzyme ever to be crystallized (1926). Finding nickel in urease’s active site (1975) was the first indication of a biological role for this metal. In this review, historical and structural features, kinetics aspects, activation of the metallocenter and inhibitors of the urea hydrolyzing activity of ureases are discussed.

The as1-casein (as1-Cas) locus in the goat is characterized by a polymorphism, the main feature of which is to be qualitative as well as quantitative. A systematic analysis performed in an autochthon southern Italy breed identified a new rare allele (M), which was characterized at both the protein and genomic level. We have compared the site-by-siteN-glycosylation status of human lactoferrin (Lf) produced in maize, a monocotyle-don, and in tobacco, usedas amodel dicotyledon.Maizeand tobacco plants were stably transformed and recombinant Lf was purified from both seeds and leaves. Phytochromes are photochromic biliproteins found in plants as well as in some cyanotrophic, photoautotrophic and heterotrophic bacteria. In many bacteria, their function is largely unknown. Here we describe the biochemi-cal and spectroscopic characterization of recombinant bacterial phyto-chrome from the opportunistic pathogen Pseudomonas aeruginosa (PaBphP). The recombinant protein displays all the characteristic features of a bonafide phytochrome.

Searching for bioactive peptides, we analyzed acidic extracts ofPhyllomedusa sauvagiiskin and found two new proteins, PSKP-1 and PSKP-2, of 6.7 and 6.6 kDa, respectively, which, by sequence homology, belong to the Kazal family of serine protease inhibitors. We cloned a cDNA for aDrosophila melanogasterhomo-logue ofmammalian heme oxygenase (HO) and constructed a bacterial expression system of a truncated, soluble form ofD. melanogasterHO (DmDHO). The purified DmDHO degraded hemin to biliverdin, CO and iron in the presence of reducing systems such as NADPH/cytochrome P450 reductase and sodium ascorbate, although the reaction rate was slower than that of mammalian HOs. Some properties of DmHO, however, are quite different from other known HOs.

DNA replication, the process of copying one double stranded DNA molecule to produce two identical copies, is at the heart of cell proliferation. This book highlights new insights into the replication process in eukaryotes, from the assembly of pre-replication complex and features of DNA replication origins, through polymerization mechanisms, to propagation of epigenetic states. It also covers cell cycle control of replication initiation and includes the latest on mechanisms of replication in prokaryotes.
