The Na+-translocating NADH:quinone oxidoreductase (Na+-NQR) from Vibrio alginolyticus was inactivated by reactive oxygen species. Highest Na+-NQR activity was observed in anaerobically prepared membranes that exhibited 1 : 1 coupling of NADH oxidation and Q reduction activities (1.6 UÆmg)1). Optical and EPR spectroscopy documented the presence of b-type cytochromes, a [2Fe)2S] cluster and an organic radical signal in anaerobically pre- pared membranes from V. alginolyticus.

Although the role of arachidonic acid (AA) in the regulation of steroidogenesis is well documented, the mechanism for AArelease isnot clear. Therefore, theaimof this studywas to characterize the role of an acyl-CoA thioesterase (ARTISt) and an acyl-CoA synthetase as members of an alternative pathway in the regulation of the intracellular levels of AA in steroidogenesis. Purified recombinant ARTISt releases AA from arachidonoyl-CoA (AA-CoA) with a Kmof 2lM.

Humanpancreatic secretions contain twomajor trypsinogen isoforms, cationic and anionic trypsinogen, normally at a ratio of 2 : 1. Pancreatitis, pancreatic cancer and chronic alcoholism lead to a characteristic reversal of the isoform ratio, and anionic trypsinogen becomes the predominant zymogen secreted. To understand the biochemical conse-quences of these alterations, we recombinantly expressed and purified both human trypsinogens and documented characteristics of autoactivation, autocatalytic degradation and Ca 2+ -dependence....

The effects of pesticide contamination on the metabolism of marine mol-luscs are poorly documented. We investigated the response of a marine bivalve, the Pacific oyster,Crassostrea gigas, using a suppression subtrac-tive hybridization method to identify up- and down-regulated genes after a 30-day exposure period to herbicides (a cocktail of atrazine, diuron and isoproturon, and to the single herbicide glyphosate). A total of 137 unique differentially expressed gene sequences was identified, as well as their asso-ciated physiological process....

It is well documented that theb-gene of the catalytic (C) subunit of protein kinase A encodes a number of splice variants. These splice variants are equipped with a variable N-terminal end encoded by alternative use of sev-eral exons located 5¢ to exon 2 in the human, bovine and mouse Cbgene.

It has been well documented that both endogenous inflammatory mediator advanced glycation end products (AGEs) and exogenous inflammatory inducer lipopolysaccharide play key roles in the initiation and development of inflammatory diseases. However, the combined inflammation-stimulatory effect of AGEs and lipopolysaccharide on endothelial cells, and, further-more, the underlying signal transduction pathways involved, have not been fully elucidated.

Apolipoprotein E is a polymorphic glycoprotein in humans with a molecu-lar mass of 34.5 kDa. It is a component of chylomicron remnants, very low density lipoprotein, low density lipoprotein and high density lipopro-tein, and is primarily responsible for maintaining plasma lipid homeostasis. In addition to these well-documented functions, recent studies in experi-mental mouse models, as well as population studies, show that apolipo-protein E also plays an important role in the development of obesity and insulin resistance. ...

Tissue transglutaminase (TG2) is a ubiquitously expressed member of the transglutaminase family of Ca 2+ -dependent crosslinking enzymes. Unlike other family members, TG2 is a multifunctional protein, which has several other well documented enzymatic and non-enzymatic functions.
