Higher life forms must have molecular mechanisms for detecting environmental information as well as mechanisms that allow for communication at the cell and tissue levels. Sensory systems detect and integrate physical and chemical information from the environment and pass this information along by the process of neurotransmission. Control and coordination of processes at the cell and tissue levels are achieved not only by neurotransmission but also by chemical signals in the form of hormones that are secreted by one set of cells to direct the activity of other cells.

Prolyl endopeptidase (PEP) is a proline-specific oligopepti-dase with a reported effect on learning and memory in dif-ferent rat model systems. Using the astroglioma cell line U343,PEP expression was reduced by an antisense technique. Measuring different second-messenger concen-trations revealed an inverse correlation between inositol 1,4,5-triphosphate [Ins(1,4,5)P3] concentration and PEP expression in the generated antisense cell lines. However,no effect on cAMP generation was observed.

Ceramide is a lipid second messenger that acts on mul-tiple-target enzymes, some of which are involved in other signal-transduction systems. We have previously demon-strated that endogenous ceramide modifies the metabolism of brain ethanolamine plasmalogens. The mechanism involved was studied. On the basis of measurements of breakdown products, specific inhibitor effects, and previ-ous findings, we suggest that a plasmalogen-selective phospholipase A2 is the ceramide target.
