Selective chemical labeling

Conventional techniques for single-base resolution mapping of epigenetic modifications of DNA such as 5-hydroxymethylcytosine (5hmC) rely on the sequencing of bisulfite-modified DNA. Here we present an alternative approach called SCL-exo which combines selective chemical labeling (SCL) of 5hmC in genomic DNA with exonuclease (exo) digestion of the bead-trapped modified DNA molecules.

The phenotypic changes in target cell induced by an exogenous agent, e.g. biologically active chemical, can be detected by labelling selected organelles with fluorescent dyes and observed under the high-throughput microscope. The changes of known biological targets (e.g. nucleus, proteins, mitochondria, microtubules, etc.) induced by a compound of interest allow predicting its mechanisms of action. High-throughput microscopy with capability of multiparametric imaging in combination with automated image-analysis is referred to as highcontent analysis or high-content screening (HCS).
