Skeletal myocytes

Skeletal muscle is one of the primary tissues involved in the development of type 2 diabetes (T2D). The close association between obesity and T2D makes it difficult to isolate specific effects attributed to the disease alone. Therefore, here we set out to identify and characterize intrinsic properties of myocytes, associated independently with T2D or obesity.

Mesenchymal stem cells are multipotent/pluripotent stromal cells that can differentiate into a diversity of cell categories depending upon the micro-environment to that they are exposed. The multipotent differentiation potential of MSCs is generally towards a constrained sum of cell types familiar to the tissue of origin and maintains the capacity for self-renewal. To assess the differentiation potential of MSCs to skeletal myocytes a male mongrel dog of 3 years was used to carry out the research.

The study was conducted on the skin of 36 goat foetus between 23 to 150 days of gestation with objective to record the sequential histological changes that take place during the differentiation of muscles in the dermis of chin region of prenatal goat. The material was divided into three groups viz. Group I (below 50 days of gestation), Group II (51-100 days of gestation) and Group III (101 days of gestation to full term) containing 12 foetii each group. The skin samples were collected and processed and stained as per standard histological technique.

The effects of physiologically relevant hypoxia on the catalytic activity of cytochromecoxidase (CytOX), mito-chondrial gene expression, and both nuclear and mito-chondrial encoded CytOX mRNA levels were investigated in murine monocyte macrophages, mouse C2C12 skeletal myocytes and rat adrenal pheochromocytoma PC12 cells. Our results suggest a coordinated down regulation of mito-chondrial genome-coded CytOX I and II and nuclear genome-codedCytOX IV andVbmRNAs during hypoxia.
