Somatic embryogenesis
-
Ebook "Plant developmental biology - Biotechnological perspectives (Volume 2)" focuses on development-related areas, including tissue culture (somatic embryogenesis, microspore embryogenesis, somaclonal variation), plant processes (photosynthesis, seed maturation and seed proteins, fatty acids, vitamins, alkaloids, flower pigments and scent), signalling (amino compound-containing lipids, auxins, cytokinins and light), and molecular genetics of developmental regulation (RNA silencing, DNA methylation, epigenetics, activation tagging, homologous recombination and synthetic promoter engineering).

Piwi-interacting RNAs (piRNAs) are a class of short (~26–31-nucleotide) non-protein-coding RNAs expressed in the metazoan germline. The piRNA pathway in arthropods is best understood in the ovary of Drosophila melanogaster, where it acts to silence active transposable elements (TEs).

Phalaenopsis is one of the most remarkable commercial genera in Orchidaceae family. Using immature explants for tissue culture of orchid results in undesired characteristics such as genetic variation and long-lasting juvenility. In this study, we optimized a protocol for somatic embryogenesis of Phalaenopsis amabilis cv. Jinan, by using mature thin cell layer (TCL) explants to minimize immature explant limitations. We examined somatic embryogenesis from two types of mature explants, viz.

The results showed that AgNPs were effective in eliminating infectious microorganisms on B. tuberous explants; which were identified included 4 species of fungi (Fusarium sp., Aspergillus aculeatus, Trichoderma sp. and Penicillium sp.) and 1 species of bacteria (Pseudomonas sp.). At concentrations of 200 ppm and 300 ppm, AgNPs were not only effective in disinfection but also increased the induction rate of somatic embryogenesis in flower stalk TCL explants (approximately 40.00%); a similar effect was observed in stem TCL explants at the same concentration.

Long non-coding RNAs (lncRNAs) are involved in variable cleavage, transcriptional interference, regulation of DNA methylation and protein modification. However, the regulation of lncRNAs in plant somatic embryos remains unclear. The longan (Dimocarpus longan) somatic embryogenesis (SE) system is a good system for research on longan embryo development.

The lower Dipteran fungus fly, Sciara coprophila, has many unique biological features that challenge the rule of genome DNA constancy. For example, Sciara undergoes paternal chromosome elimination and maternal X chromosome nondisjunction during spermatogenesis, paternal X elimination during embryogenesis, intrachromosomal DNA amplification of DNA puff loci during larval development, and germline-limited chromosome elimination from all somatic cells.

Somatic embryogenesis receptor-like kinases (SERKs) are leucine-rich repeat receptor-like kinases associated with various signaling pathways. These kinases have a relationship with stress signals, and they are also believed to be important for regulating plant growth. However, information about this protein family in apple is limited.

Coffee production relies on plantations with varieties from Coffea arabica and Coffea canephora species. The first, the most representative in terms of coffee consumption, is mostly propagated by seeds, which leads to management problems regarding the plantations maintenance, harvest and processing of grains.

Somatic embryogenesis (SE) is a process of somatic cells that dedifferentiate to totipotent embryonic stem cells and generate embryos in vitro. Longan SE has been established and wildly used as model system for studying embryogenesis in woody plants, SE-related genes had been characterized.

In citrus, genetic improvement via biotechnology is hindered by the obstacle of in vitro regeneration via somatic embryogenesis (SE). Although a few B3 transcription factors are reported to regulate embryogenesis, little is known about the B3 superfamily in citrus, and which members might be involved in SE.

Eucalyptus, a highly diverse genus of the Myrtaceae family, is the most widely planted hardwood in the world due to its increasing importance for fiber and energy. Somatic embryogenesis (SE) is one large-scale method to provide commercial use of the vegetative propagation of Eucalyptus and dedifferentiation is a key step for plant cells to become meristematic.

Somatic embryogenesis (SE) is a promising technology for plant vegetative propagation, which has an important role in tree breeding. Though rubber tree (Hevea brasiliensis Muell. Arg.) SE has been founded, few late SE-related genes have been identified and the molecular regulation mechanisms of late SE are still not well understood.

Somatic embryogenesis (SE) is the process in which somatic embryos develop from somatic tissue in vitro on medium in most cases supplemented with growth regulators. Knowledge of genes involved in regulation of initiation and of development of somatic embryos is crucial for application of SE as an efficient tool to enable genetic improvement across genotypes by clonal propagation.

Somatic embryogenesis in grapevines is a complex process that depends on many physiological and genetic factors. One of its main limitations is the process of precocious germination of the somatic embryos in differentiation medium.

Arabinogalactan-proteins (AGPs) are a class of hydroxyproline-rich proteins (HRGPs) that are heavily glycosylated (> 90%) with type II arabinogalactans (AGs). AGPs are implicated in various plant growth and development processes including cell expansion, somatic embryogenesis, root and stem growth, salt tolerance, hormone signaling, male and female gametophyte development, and defense.

Genotype independent transformation and whole plant regeneration through somatic embryogenesis relies heavily on the intrinsic ability of a genotype to regenerate. The critical genetic architecture of non-embryogenic callus (NEC) cells and embryogenic callus (EC) cells in a highly regenerable cotton genotype is unknown.

Coconut (Cocos nucifera L.), is an important perennial crop cultivated across the globe. The demand for quality planting materials is increasing across the world for the establishment of new orchards, rejuvenation of existing orchards and production of quality nuts. Plant tissue culture is an alternate approach for the production of quality planting material. In this investigation, influence of auxin and cytokinin on somatic embryogenesis in coconut is studied using sliced mature embryos as explants in modified Y3 media supplemented with various concentrations of 2,4-D and kinetin.

Five maize lines prevalently used by breeders in the maize program were screened for their ability to form type II callus. Mature seeds were used as explant for in vitro culturing. One inbred line showed high embryogenic response and formed friable, type II calli and also exhibited successful plant regeneration. Ten culture media compositions based on Murashige and Skoog (1962) salts thus investigated differed with respect to the combinations of concentrations of auxins (IBA, NAA, picloram, 2, 4-D) and cytokinins (BAP, kinetin) were used.

Chrysanthemum is a second most important flower crop globally. Marigold is a introduced cultivar gaining popularity among farmers and consumers of southern part of India due to its colour and high shelf life. Due to its commercial importance as quality planting material and cut flowers it is difficult to mitigate the gradually increasing demand by only vegetative means of propagation which is time consuming with low multiplication rate. Plant tissue culture is a better alternative tool to overcome conventional limitations.
