The present in vitro study was conducted with aim to know the antifungal activity of three bio-agents (Trichoderma viride, Pseudomonas fluorescens and Bacillus subtilis), Neem oil and amendments (FYM and Neem cake) against Sclerotium rolfsii causing Stem rot in groundnut. The study was conducted in Plant Pathology laboratory, SHUATS during 2020.

Stem and pod rot of groundnut caused by the pathogen Sclerotium rolfsii, which has a wide host range and it affects the yield losses up to 30-40% in groundnut in India. In the present investigation efforts were made to test the influence of soil moisture on the viability of sclerotium of groundnut stem rot causing pathogen Sclerotium rolfsi. Based on the results least sclerotial germination (76.66) occurred at soil pH 9.0 and this indicates that basic pH is less favorable for sclerotial germination. Moisture levels of 50 per cent (control) and 40 per cent were found to be best for sclerotial germination (100%).

Genetic variability among the 24 isolates of S. rolfsii was studied by using molecular markers like ITS-PCR and RAPD primers. Amplification of ITS region of rDNA with specific ITS1 and ITS4 universal primers produced approximately 650 to 700 bp in all the isolates of the fungus confirmed that all the isolates obtained are Sclerotium rolfsii and were sequenced. Identity of the isolates was confirmed with sequences of NCBI data base of S. rolfsii.

Stem rot of groundnut (Arachis hypogaea L.) caused by Sclerotium rolfsii Sacc. is a serious fungal disease of groundnut. Conventional uses of fungicides are toxic to environment thus an eco-friendly new approach of management with nonconventional chemicals as seed treatment was undertaken. Seeds of groundnut were soaked in non-conventional chemicals namely, ZnSO4, ZnCl2, BaSO4, BaCl2, Cycocel and Indole Acetic Acid (I.A.A) @ 10-4 and 10 -5 Molar concentrations for 24 hours to evaluate its bio-efficacy against the disease.

Groundnut (Arachis hypogaea L.) is an important edible leguminous oilseed crop. Diseases of groundnut act as the limiting factor to its economic production. Recently, the stem rot caused by Sclerotium rolfsii is becoming severe disease of groundnut in India. Crop Losses up to 59 per cent due to stem rot disease have been reported by many researchers.

Groundnut is a major legume and important oil seed crop belongs to family Fabaceae. A large number of diseases attack groundnut plant in India, of these stem rot caused by Sclerotium rolfsii is the most common disease. The present study was conducted in experimental field of SHUATS for management of stem rot disease of groundnut (Sclerotium rolfsii) which is most important destructive disease of groundnut in India.
