An optimization study of an integrated periodic counter-current chromatography (PCC) process in a monoclonal antibody (mAb) downstream process at lab scale, is presented in this paper. The optimization was based on a mechanistic model of the breakthrough curve in the protein-A capture step.

The Obscure Features Hypothesis (OFH) for innovation states that a two-step process undergirds almost all innovative solutions: (1) notice an infrequently observed or new (i.e., obscure) feature of the problem and (2) construct an interaction involving the obscure feature that produces the desired effects to solve the problem. The OFH leads to a systematic derivation of innovation-enhancing techniques by engaging in two tasks. First, we developed a 32-category system of the types of features possessable by a physical object or material.

A cost-effective approach to the diagnostic evaluation of delirium allows the history and physical examination to guide tests. No established algorithm for workup will fit all delirious patients due to the staggering number of potential etiologies, but one step-wise approach is detailed in Table 26-3. If a clear precipitant is identified early, such as an offending medication, then little further workup is required. If, however, no likely etiology is uncovered with initial evaluation, an aggressive search for an underlying cause should be initiated.
