  • In this study, the effect of plant growth regulators (BA and NAA), ripe bananas in combination with potato, humic acid, and silver nanoparticles (AgNPs) were investigated to find a suitable condition for in vitro plant regeneration from R. imschootiana protocorm-like body (PLB). The results showed that after 45 days of culture, MS medium supplemented with 100 g ripe banana/liter and 100 g potato/liter was suitable for regenerating shoots from the protocorm-like body (PLB) (17.56 shoots/PLBs and 100.00% of shoot-regenerating PLBs).

  • An efficient regeneration protocol was established for a rare plant species, Begonia coptidifolia. Using leaf explants, adventitious shoots were induced on Murashige and Skoog (MS) basal medium using various concentrations (1.0–10.0 mM) of three cytokinins, 6-benzyladenine (BA), thidiazuron (TDZ) and kinetin (KIN). However, adventitious shoots could not be induced on MS medium supplemented with auxins [α-naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D)] tested in the 1.0–10.0 mM range.

  • Roses are one of the most popular fl owers in the world because of their beauty, color, aroma as well as their various uses and Red Eden rose is one of the most popular hybrid roses. Results of the study showed the most optimal medium for shoot multiplication, which was MS + 30 g/l of sucrose + 8 g/l of agar supplemented with 0.5 mg/l of BA. The optimal medium for growth and fl owering was ½MS + 30 g/l of sucrose + 8 g/l of agar supplemented with 30 μM of AgNO3 . However, NAA at all tested concentrations did not show any eff ect on in vitro plant growth.

  • Hydrangea (Hortensia) is a highly popular ornamental plant for garden decoration, and now it is commercially produced for cut flower branches. The aim of this study was to set up a protocol for large scale for in vitro propagation of Hydrangea macrophylla, for this purpose two type of explant (shoot tip and node) were sterilized and the best percentage of survival explants (40%) was obtained when explants immersed for 20 min in 1.0 or 1.5 % NaOCl. For in vitro multiplication, Murashige and Skoog medium supplemented with 1.0 mg/l BA and 2.0 mg/l Kin was the best treatment on shoot number.

  • The aim of this study was to isolate and optimize the growth conditions of thermophilic microorganism from hot springs. The isolation was conducted by using the mineral salt basal medium supplemented with 0.6% yeast extract at 500C.   • Double sterilized and pre-chilled shoot apices of Galmast cultivar obtained from mature trees (MTSTs) and in vitro raised seedlings (SBSTs) were inoculated on MS medium augmented with BA, Kn and TDZ with and without PG. Increase in BA concentration up to 5.0µM resulted in axillary shoot proliferation in about 40% shoots with a maximum of 14±0.72 shoots per explant.   • Violar tricolor belongs to Viola genus, Violaceae family, Violar tricolor plant is not only grown as an ornamental plant, but also a medicinal herb. In this study, some of factors affect on the process of micropropagation were investigated. In the experiment of sterilization explants, seed germination and shoot formation ability of the Violar tricolor L. reached the best result when they were sterilized by 0.1% HgCl2 for 1 minute.

  • An in vitro protocol for plant regeneration through organogenesis was established for Picrorhiza kurroa Royle Ex. Benth (Scrophulairaceae). The plant is a principle source of glycoside that is Picroside-I, Picroside-II and Kutkoside.   • A study on the development of in vitro propagation protocol for newly developed mutants of Carnation (Dianthus caryophyllus L.)” was carried out during 2017-2018. Results reveal that all the mutants responded well to the in vitro propagation, however ‘UHFS Car-4’ exhibited best propagation potential. Calcium Hypochlorite (5%) treatment for 20 min. is suggested for surface sterilization of explants as it gives 100% uncontaminated growing cultures. For initial culture establishment, MS medium supplemented with 2.0 mgl-1 BA proved to be most effective resulting in earliest establishment.

  • The results showed that protocorms were formed from seeds culture on the ½ MS medium supplemented with 10% potatoes extract within 8 weeks. Protocorms which were cultured on the ½ MS medium containing 30 g/l sucrose, 0.5 g/l AC, 7 g/l agar, 0.1 mg/l NAA and 1 mg/l BA were optimal for PLB formation (8.67 PLBs/sample) after 8 weeks culture.   • A method for micropropagation was developed for Dendrocalamus asper, an economically and environmentally important bamboo. Disinfected seeds were cultured in flasks containing 20 ml of Murashige and Skoog’s medium (MS) supplemented with BA (1.0-7.0 mg l-1 ) or kinetin (1.0-7.0 mg l-1 ). Multiple shoots (6.53) were formed on MS medium supplemented with 3.0 mg l-1 BA and reached 1.49 cm in length. Continuous shoot proliferation was achieved on a MS medium supplemented with BA (1.0-7.0 mg l -1 ). The multiplication rate of 3.30 fold was achieved on MS medium supplemented with 3.0 mg l-1 BA.

  • We report here the protocol for Zedoary (Curcuma zedoaria Roscoe) callus and cell suspension cultures. The MS medium supplemented with 2% sucrose, 1.0 mg/l 2,4-D and 1.0 mg/l BA was effective for callus induction from in vitro leaf-base explants of Zedoary. During subcultures, secondary proliferated calli were subsequently produced from initial induced calli on the MS medium with 0.5 mg/l 2.4-D and 0.5 mg/l BA. These calli were light yellow in color, compact and friable.
