The interaction between the c11ring and thececomplex, forming the rotor of theIlyobacter tartaricus ATP synthase, was probed by surface plasmon resonance spectroscopy andin vitro reconstitution analysis. The results pro-vide, for the first time, a direct and quantitative assessment of the stability of the rotor. The data indicated very tight binding between the c11ring and the cecomplex, with an apparent Kd value of approximately 7.4 nm.

Titin is known to interact with actin thin filaments within the I-band region of striated muscle sarcomeres. In this study, we have used a titin fragment of 800 kDa (T800) purified from striated skeletal muscle to measure the effect of this interaction on the functional properties of the actin– myosin complex. MALDI-TOF MS revealed that T800 contains the entire titin PEVK (Pro, Glu, Val, Lys-rich) 1 domain.

N-Glycosylation is a cotranslational and post-translational process of pro-teins that may influence protein folding, maturation, stability, trafficking, and consequently cell surface expression of functional channels. Here we have characterized two consensus N-glycosylation sequences of a voltage-gated K + channel (Kv3.1). Glycosylation of Kv3.1 protein from rat brain and infected Sf9 cells was demonstrated by an electrophoretic mobility shift assay.

The interaction of calreticulin with mannan-binding lectin (MBL) was investigated by solid-phase binding assays.
