Two ORFs, cphA and cphB, encoding proteins CphA and CphB with strong similarities to plant phytochromes and to the cyanobacterial phytochrome Cph1 of Synechocystis sp. PCC 6803 have been identified in the filamentous cyanobacterium Calothrix sp. PCC7601. While CphA carries a cysteine within a highly conserved amino-acid sequence motif, to which the chromophore phytochromobilin is covalently bound in plant phytochromes, in CphB this position is changed into a leucine.

This studywas designed to yield data on the supramolecular organization of the phycobilisome apparatus from Synechocystis, and the possible effects of environmental stress on this arrangement. Phycobilisomes were dissociated ina low ionic strength solutionandaquantitative estimation of the protein components present in each subcomplex was obtained using liquid chromatography coupled on-line with a mass spectrometer equipped with an electrospray ion source (ESI-MS).

The PsbH protein, a small subunit of the photosystem II complex (PSII),was identi®edas a6-kDaproteinband in the PSII core and subcore (CP47±D1±D2±cytb-559) from the wild-type strain of the cyanobacteriumSynechocystisPCC 6803. The protein was missing in the D1±D2±cyto-chromeb-559 complex and also in all PSII complexes isolated from IC7, a mutant lacking the psbHgene.

Genes homologous to hydrogenase accessory genes are scattered over the whole genome in the cyanobacteriumSynechocystissp. PCC 6803. Deletion and insertion mutants of hypA1 (slr1675), hypB1 (sll1432), hypC, hypD, hypE and hypF were constructed and showed no hydrogenase activity. Involvement of the respective genes in maturation of the enzyme was con-firmed by complementation. Deletion of the additional homologueshypA2 (sll1078) and hypB2(sll1079) had no effect on hydrogenase activity. Thus, hypA1andhypB1are specific for hydrogenase maturation....

Two isoforms of a heme oxygenase gene,ho1andho2, with 51% identity in amino acid sequence have been identified in the cyanobacterium Synechocystissp. PCC 6803. Isoform-1, Syn HO-1, has been characterized, while isoform-2, Syn HO-2, has not. In this study, a full-lengthho2gene was cloned using synthetic DNA and Syn HO-2 was demonstrated to be highly expressed inEscherichia colias a soluble, catalytically active protein.

Heme oxygenase (HO) catalyzes the oxidative degradation of heme utilizing molecular oxygen and reducing equiva-lents. In photosynthetic organisms, HO functions in the biosynthesis of such open-chain tetrapyrroles as phyto-chromobilin and phycobilins, which are involved in the sig-nal transduction for light responses and light harvesting for photosynthesis, respectively.

PCC6803, were predicted for the whole genome sequence by estimating changes in the binding free energy (DDG A total ) for SYCRP1 for those sites. In order to identify an enzyme capable of Fenton reaction inSynechocystis, we purified an enzyme catalyzing one-electron reduction oft-butyl hydro-peroxide in the presence of FAD and Fe(III)-EDTA. The enzyme was a 26 kDa protein, and its N-terminal amino acid sequencing revealed it to be DrgA protein previously reported as quinone reductase [Matsuo M, Endo T and Asada K (1998)Plant Cell Physiol39,751–755].

The charged quaternary ammonium compounds – methyl, ethyl and benzyl viologens – generate reactive oxygen species in photosynthetic cells. Three independent methyl viologen-resistant spontaneous mutants of Synechocys-tis sp. PCC 6803 were identified, in which the conserved R115 residue of the Slr1174 protein was replaced with G115, L115 and C115. It has been shown that the Synechocystis cytochrome P450 enzyme CYP120A1 is a retinoic acid-binding polypeptide. In this work, we determined the reaction catalyzed by CYP120A1 and investigated its substrate specificity in vitro.
