The metalloproteinase pregnancy-associated plasma pro-tein-A (PAPP-A) cleaves a subset of insulin-like growth factor binding proteins (IGFBP), which inhibit the activities of insulin-like growth factor (IGF). Through this proteolytic activity, PAPP-A is believed to regulate IGF bioavailability in several biological systems, including the human repro-ductive system and the cardiovascular system. PAPP-A adheres to mammalian cells by interactions with glycos-aminoglycan (GAG), thus targeting the proteolytic activity of PAPP-A to the cell surface. ...

CCN members have typical structures composed of four conserved cysteine-rich modules and their variants lack-ing certain modules, generated by alternative splicing or gene mutations, have been described in various pathological conditions.

The kinase noncatalytic C-lobe domain (KIND) is a putative protein–protein interaction module. Four KIND-containing proteins, Spir-2 (actin-nuclear factor), PTPN13 (protein tyrosine phosphatase), FRMPD2 (scaffold protein) and very-KIND (v-KIND) (brain-specific Ras guanine nucleotide exchange factor), have been identified to date.
