Two cysteine residues were introduced to selected positions (Serine-309 and Serine-43) of the Thermoplasma acidophilum citrate synthase. Serine-309 Cysteine mutation drastically reduced the activity, as well as the thermostabiity of the enzyme.

The 26S proteasome plays a critical role in many cellular processes by degrading a wide range of both short-lived and long-lived proteins. In this review, proteasome structure and the mechanisms of substrates targeting to the 26S proteasome are described.

A gene with significant similarity to bacterial Lon proteases was identified during the sequencing of the genome of the thermoacidophilic archaeon Thermoplasma acidophilum. Protein sequence comparison revealed thatThermoplasma Lon protease (TaLon) ismore similar to theLonBproteases restricted to Gram-positive bacteria than to the widely dis-tributed bacterial LonA.

Members of the 2-oxoacid dehydrogenase multienzyme complex family play a key role in the pathways of central metabolism. Post-translational lipoylation of the dihydrolipoyl acyltransferase component of these com-plexes is essential for their activity, the lipoyllysine moiety performing the transfer of substrates and intermediates between the different active sites within these multienzyme systems.
