  • Activated charcoal (AC) is highly adsorbent and is often used to promote seedling growth in plant tissue culture; however, the underlying molecular mechanism remains unclear. In this study, root and leaf tissues of 10-day-old seedlings grown via immature embryo culture in the presence or absence of AC in the culture medium were subjected to global transcriptome analysis by RNA sequencing to provide insights into the effects of AC on seedling growth.

  • Pre-requisite for any plant biotechnological approach is development of an efficient plant regeneration system in that crop. In this regard the in vitro regeneration was achieved by using cotyledon explants from aseptically raised seedlings of popular Capsicum F1 hybrids Bharat and Indra. Seeds of both hybrids were exposed to different treatments for proper germination, were decontaminated and sown in vitro on half-strength MS medium.

  • Hydrangea (Hortensia) is a highly popular ornamental plant for garden decoration, and now it is commercially produced for cut flower branches. The aim of this study was to set up a protocol for large scale for in vitro propagation of Hydrangea macrophylla, for this purpose two type of explant (shoot tip and node) were sterilized and the best percentage of survival explants (40%) was obtained when explants immersed for 20 min in 1.0 or 1.5 % NaOCl. For in vitro multiplication, Murashige and Skoog medium supplemented with 1.0 mg/l BA and 2.0 mg/l Kin was the best treatment on shoot number.

  • Pomegranate can be propagated by seed, air layering, grafting, shoot cuttings and tissue culture. Seed propagation is used to raise the rootstocks that can be utilized for germplasm conservation and rootstocks breeding purpose. Vegetative propagation methods are used to multiply selected genotypes that bring similar character in form of clones having similar characters to mother plant. Vegetative propagation techniques (hard wood cutting, layering) are practices to raise the saplings for commercial production in pomegranate, because it perpetuates all characteristics of each cultivar.

  • Some glycosidic enzymes secreted by the gut of Eudrilus eugeniae and the abundance of bacteria producing similar enzymes in the gut were studied in order to know the extent of enzymatic contribution by the worm gut and the ingested bacteria. 120 different bacteria were isolated from the worm gut and their enzymatic activities were detected by using respective media and reagents. In vitro gut wall tissue culture of various gut portions was done to assess enzymatic activities in gut wall and in the culture medium.

  • Shoot apices of Physalis minima L. were cultured on fifteen full strength MS medium supplemented with various concentrations of cytokinins (BAP, KIN and TDZ). Tissue culture responses namely elongation of existing shoot and differentiation of multiple shoots were observed from the cultured explants. The establishment of cultured explants was not affected by the cytokinin concentration in the medium but overall shoot apices showed better establishment on medium supplemented with BAP than other cytokinins.

  • The low cost alternative components of tissue culture media were evaluated for their potential to supply nutrients and to support growing explant of Banana cultivar in vitro for their cost effectiveness against conventional high cost media. Experiment was laid out using completely randomized design (CRD) with ten replications. Six culture media viz.   • The present study was carried out to monitor somaclonal variations of banana cv. Ney Poovan (AB) during micropropagation using random amplified polymorphic DNA (RAPD). Sword suckers were used as explants and regenerated on MS media supplemented with BAP alone, BAP + Kinetin and TDZ at varying levels of concentrations. The regenerated shoots were again subcultured on MS medium fortified with plant growth regulators at different concentrations. Rooting was obtained on ½ MS medium supplemented with 2.00 mg/l IBA and 2 g/l of activated charcoal.

  • In the present investigation, a simple, rapid and efficient in vitro regeneration system was developed in green gram (Vigna radiata L. Wilczek) cv. IPM-02-03 using cotyledonary explants excised from a 4 day old seedling.
