Prolyl endopeptidase (PEP) is a proline-specific oligopepti-dase with a reported effect on learning and memory in dif-ferent rat model systems. Using the astroglioma cell line U343,PEP expression was reduced by an antisense technique. Measuring different second-messenger concen-trations revealed an inverse correlation between inositol 1,4,5-triphosphate [Ins(1,4,5)P3] concentration and PEP expression in the generated antisense cell lines. However,no effect on cAMP generation was observed.

To learn about the correlation between allostery and ligand binding of the Tet repressor (TetR) we analyzed the effect of mutations in the DNA read-ing head–core interface on the effector specific TetR i2 variant. The same mutations in these subdomains can lead to completely different activities, e.g. the V99G exchange in the wild-type leads to corepression by 4-ddma-atc without altering DNA binding. Prolyl oligopeptidase (POP) is a serine protease that cleaves small peptides at the carboxyl side of an internal proline residue. Substance P, arginine– vasopressin, thyroliberin and gonadoliberin are proposed physiological substrates of this protease.
