As with other complex human diseases, risk in coronary disease loci is determined primarily by altered expression of the causal gene, due to variation in binding of transcription factors and chromatin-modifying proteins that directly regulate the transcriptional apparatus.

Gonadal sex differentiation proceeds by the interplay of various genes including the transcription factors and secretory factors in a complex network. Dmrt1 and Sox9 were considered to be the essential genes involved in the testicular differentiation in mammals, birds, reptiles, and fishes. Here, we have successfully isolated and characterized cDNAs encoding the Rana chensinensis orthologs of Dmrt1 and Sox9, and we have determined patterns of expression during gonadal differentiation and adult tissues.

CBLs can perceive and bind Ca2+ under adversity and then specifically interact with CIPKs. The activated CBL–CIPK complex phosphorylates downstream target proteins (such as ion channels and transporters), or regulates transcription factors and stress responsive genes.

Recently, boron (B) has been found to form a complex with anthocyanin, which could be evidence for the B tolerance mechanisms that reduce free B in the leaf tissues of plants. However, the molecular mechanism of anthocyanin biosynthesis and transport has not been satisfactorily elucidated in plants exposed to toxic B. Therefore, the changes in expression levels of some of the phenylpropanoid pathway genes, early and late flavonoid biosynthetic genes, and transcription factors related to anthocyanin biosynthesis and transport were determined in Arabidopsis thaliana under B toxicity.

Precise regulation of gene expression is required for embryonic stem cell (ESC) differentiation. Transcription factor (TF) networks coordinate the balance of pluripotency and differentiation in response to extracellular and intracellular signals. Chromatin factors work alongside TFs to achieve timely regulation of gene expression for differentiation process. Our previous studies showed that a member of the Sin3a corepressor complex, Arid4b, is critical for proper mouse ESC differentiation into mesoderm and endoderm.

RNA polymerase II (Pol II) is a 12 subunit protein complex from yeast to human that is required for gene expression. Gdown1 containing Pol II [Pol II (G)] is a special form of Pol II that is catalytically inactive and heavily depends on the 30-subunit Mediator complex for its activator and basal dependent function in vitro. Here we report for the first time, the identification and the generation of a 15-subunit human Mediator complex via the novel multibac baculovirus expression system that is fully responsive to Pol II (G).

Fruit ripening is an intricate developmental process driven by a highly coordinated action of complex hormonal networks. Ethylene is considered as the main phytohormone that regulates the ripening of climacteric fruits. Concomitantly, several ethylene-responsive transcription factors (TFs) are pivotal components of the regulatory network underlying fruit ripening. Topological association domains (TADs) are chromosomal domains characterised by frequent internal DNA-DNA interactions. The transcription factor CTCF binds to conserved DNA sequence patterns called CTCF binding motifs to either prohibit or facilitate chromosomal interactions. TADs and CTCF binding motifs control gene expression, but they are not yet well defined in the bovine genome. In this paper, we sought to improve the annotation of bovine TADs and CTCF binding motifs, and assess whether the new annotation can reduce the search space for cis-regulatory variants.

Brassica crops are cultivated widely for human consumption and animal feed purposes, and oilseed rape/canola (Brassica napus and rapa) is the second most important oilseed worldwide. n the present work, a pioneering attempt has been made to overexpress master regulator binary transcription factor complex formed by HlWRKY1 and HlWDR1 using a plant expression vector to enhance the level of prenylflavonoid and bitter acid content in the hop. Subsequently, we performed transcriptional profiling using high-throughput RNA-Seq technology in leaves of resultant transformants and wild-type hop to gain in-depth information about the genome-wide functional changes induced by HlWRKY1 and HlWDR1 overexpression.
