Transgenic mice

To facilitate studies on di€erentiation of adrenocortical cells and regulation of steroidogenic genes, we established cell lines from adrenals of adult transgenic mice harboring a temperature-sensitive large T-antigen gene of simian virus 40. Adrenal glands of the mice exhibited normal cortical zonation including a functionally undi€erentiated cell-layer between the aldosterone-synthesizing zona glomerulosa cells and the corticosterone-synthesizing zona fasciculata cells.

Class II MHC genes (for example, the humanHLA-DRAgene) are expressed at the cell surface in many professional and nonprofessional anti-gen-presenting cells in a variety of anatomical locations. Loss of cannabinoid receptors (CB1) occurs prior to neuro-degeneration in Huntington’s disease (HD). The levels and distribution of CB1 RNA were equivalent in 3-week-old mice regardless of genotype demonstrating that the specific factors and appropriate chromatin structure that lead to the transcription ofCB1were present in the striatum of young R6/2 and R6/1 transgenic HD mice.

We previously identified a partial expressed sequence tag clone corresponding toNARG2in a screen for genes that are expressed in developing neurons and misexpressed in transgenic mice that lack functional N-methyl-D-aspartate receptors. Here we report the first characterization of the mouse and humanNARG2genes, cDNAs and the proteins that they encode.Mouse andhumanNARG2 consist of 988 and 982 amino acids, respectively, and share 74% identity.

Lactoferrin (LF) is an iron-binding glycoprotein of the innate host defence system. To elucidate the role ofN-linked glycosylation in protection of LF against proteolysis, we compared the tryptic susceptibility of human LF (hLF) variants from human milk, expressed in human 293(S) cells or in the milk of transgenic mice and cows. The analysis revealed that recombinant hLF (rhLF) with mutations Ile130fiThr and Gly404fiCys was about twofold more susceptible than glycosylated and unglycosylated variants with the naturally occurring Ile130 and Gly404....

Intracellularly expressed antibodies (intrabodies) have been used to inhibit the function of various kinds of protein inside cells. However, problems with stability and functional expression of intrabodies in the cytosol remain unsolved. Huntington’s disease (HD) is a progressive neurodegenerative disorder characterized by multifarious dysfunctional alterations including mitochon-drial impairment. In the present study, the formation of inclusions caused by the mutation of huntingtin protein and its relationship with changes in energy metabolism and with pathological alterations were investigated both in transgenic and 3-nitropropionic acid-treated mouse models for HD.

Here we report on the role of Glu59 in the fumarate-mediated allosteric regulation of the human mitochondrial NAD(P) + -dependent malic enzyme (m-NAD-ME). In the present study, Glu59 was substituted by Asp, Gln or Leu. Our kinetic data strongly indicated that the charge properties of this residue significantly affect the allosteric activation of the enzyme.

The betulinic acid (BetA) purified fromPulsatilla chinensis(PC) has been found to have selective inhibitory effects on hepatitis B virus (HBV). In hepatocytes from HBV-transgenic mice, we showed that BetA substantially inhibited HBV replication by downregulation of manganese superoxide dismutase (SOD2) expression, with subsequent reactive oxygen species gen-eration and mitochondrial dysfunction.
