The aim of this study was to evaluate the efficacy of the p53-reactivating drugs RITA and nutlin3a in killing myeloma cells. Methods: A large cohort of myeloma cell lines (n = 32) and primary cells (n = 21) was used for this study. This cohort contained cell lines with various TP53 statuses and primary cells with various incidences of deletion of chromosome 17.

Tumor suppressor protein p53 plays a central role in maintaining genomic integrity [1–3]. In unstimulated cells p53 is maintained at very low levels but in response to DNA damage or other stress stimuli, such as hypoxia or activation of oncogenes, p53 becomes stabilized and accumulates in the cell [4, 5]. p53 can exert its tumor suppressor function in different ways (Fig. 1.1).
