An improved reverse genetics system for influenza A virus generation and its implications for vaccine production present an improved system for the generation of influenza viruses that allows the easy and repro ducible production of vaccine viruses in vero cells. Application of this system may be especially advantageous in situations of outbreaks of highly pathogenic avian influenza viruses.

Keeping poultry makes a substantial contribution to household food security throughout the developing world. It helps diversify incomes and provides quality food, energy, fertilizer and a renewable asset in over 80 percent of rural households. Prion diseases are fatal neurodegenerative disorders caused by proteina-ceous infectious pathogens termed prions (PrP Sc ). To date, there is no pro-phylaxis or therapy available for these transmissible encephalopathies. Passive immunization with monclonal antibodies recognizing the normal host-encoded prion protein (PrP C ) has been reported to abolish PrP Sc infec-tivity and to delay onset of disease.

An illuminating example is the case of Rotashield, a rhesus reassortant rotavirus vaccine, which was introduced for routine use in the United States in the late 1990s. Within 9 months of its introduction, cases of intussusception were reported by the CDC to be temporally associated with the administration of the initial vaccine dose. Risk Assessment Vaccines are considered safe when the risk of use is judged to be acceptable in relation to the benefits. For vaccines given to healthy individuals for diseases that are no longer common, acceptable risks are set at very low levels—indeed, far lower than for most medical products. The Immune Response While many constituents of infectious microorganisms and their products (e.g., exotoxins) are or can be rendered immunogenic, only some stimulate protective immune responses that can prevent infection and/or clinical illness or (as in the case of rotavirus) can attenuate illness, providing protection against severe disease but not against infection or mild illness. The immune system is complex, and many factors—including antigen composition and presentation as well as host characteristics—are critical for stimulation of the desired immune responses (Chap. 308).
