Simmondsia chinensis (jojoba) is the only plant known to store wax esters instead of triacylglycerols in its seeds. Wax esters are composed of very-long-chain monounsaturated fatty acids and fatty alcohols and constitute up to 60% of the jojoba seed weight.

Waxes are esters of long-chain fatty acids with long-chain fatty alcohols found abundant in the environment in various forms. Bacteria degrading these waxes in agricultural residues, soils and insects are gaining increased attention. In India, about 500 million tonnes of crop residues are generated annually and these crop residues are rich in cellulose, hemicellulose, lignin, pectin, cutin, waxes, fats and oils. Accumulation of waxy rich crop residues in the environment decreases the rate of decomposition and increase the hydrophobicity of soils.

Very long chain fatty acids (C20 and longer) are synthesized via fatty acid elongases at ER. Plastid generated C16 and C18 fatty acids and malony CoA are substrates for these elongases. The cuticular waxes are a mixture of mostly aliphatic very long chain fatty acids (VLCFA) and variable amount of triterpenoids and phenyl propanoids. Cuticle is composed of inter-esterification of ω and mid chain hydroxy and epoxy C16 and C18 fatty acids and glycerol, and cuticular waxes.

The direction of science is often driven by methodological progress, and the topic of this book is no exception. I remember sitting with a visitor on the terrace of a hotel overlooking Lake Constance in the early 1970s. We were discussing the gravimetric method of measuring total lipids in zooplankton. A few years later, as a visitor in Clyde E. Goulden’s lab, I was greatly impressed by the ability of an instrument called an Iatroscan to discriminate and quantify specific lipid classes (e.g., triacylglycerols, polar lipids, wax esters).
