Culture of clinical samples is the confirmatory test for the diagnosis of paratuberculosis but, it is time consuming due to its long incubation period, further it requires a skilled personnel to monitor the growth. Use of molecular techniques such as PCR is advantageous over the long incubation period in culturing of samples; more than eight months of incubation is needed to declare a sample to be negative.

The bacteriological diagnosis of tuberculosis (TB) is largely dependent on the Ziehl-Neelsen (ZN) microscopy. This method has a low sensitivity, which can be improved by the concentration of the specimen with sodium hypochlorite or Bleach (NaOCl), followed by sedimentation after doing LED. This study was conducted to evaluate the efficiency of bleach sedimentation technique in detection of Mycobacterium tuberculosis by LED microscopy. A Total of 100 sputum samples from the patients who visited RNTCP of Rama Medical College Hospital and Research Centre between April 2015 and March 2016.

Johne’s disease (Paratuberculosis), a debilitating chronic granulomatous enteritis of domesticated and wild ruminants that causes huge economic and production losses to the dairy farmers. A six-year-old Jersey crossbred cow was presented with a history of chronic diarrhoea, gradual weight loss, reduction in milk yield and poor response to therapy. Based on history, clinical examination, ZiehlNeelsen staining and IS900 polymerase chain reaction of the faeces the case was diagnosed as Johne’s disease.
