Ebook "Plant ABC transporters" is devoted to the fascinating superfamily of plant ATP-binding cassette (ABC) transporters and their variety of transported substrates. It highlights their exciting biological functions, covering aspects ranging from cellular detoxification, through development, to symbiosis and defense. Moreover, it also includes a number of chapters that center on ABC transporters from non-Arabidopsis species.

We performed an in-depth analysis of the ABC gene family in Aedes aegypti (Diptera: Culicidae), which is an important vector species of arthropod-borne viral infections such as chikungunya, dengue, and Zika. Despite its importance, previous studies of the Arthropod ABC family have not focused on this species.

The ATP-binding cassette (ABC) transporter superfamily is comprised predominantly of proteins which directly utilize energy from ATP to move molecules across the plasma membrane. Although they have been the subject of frequent investigation across many taxa, arthropod ABCs have been less well studied.

Cystic echinococcosis (CE) is a life-threatening zoonosis caused by the larval form of Echinococcus granulosus tapeworm. Our previous study showed that an approved drug pyronaridine (PND) is highly effective against CE, both in vitro and in an animal model.

Transportation of pigment precursors or related metabolites from cytoplasm to subcellular pigment granules is one of the key steps in insect pigmentation and usually executed via such transporter proteins as the ATP-binding cassette (ABC) transmembrane transporters and small G-proteins (e.g. Rab protein).

The growth- and plasticity-associated protein-43 (GAP43) is biasedly expressed in indigestive system and nervous system. Recent study has shown that GAP43 is responsible for the development of neuronal growth and axonal regeneration in normal nervous tissue, while serves as a specific biomarker of relapsed or refractory neuroblastoma.

Efforts are in progress to conserve critically endangered Gyps spp. vultures by identifying and removing the vulture-unsafe drugs from the food chain. Several drugs namely diclofenac, aceclofenac, ketoprofen and nimesulide are found to cause hyperuricemia and fatal nephrotoxicity in Gyps vultures. Research on development of an in vitro model for screening drugs in practice and the new molecules for their hyperuricemic potential can facilitate the conservation efforts.

ATP-binding cassette proteins have been recognized as playing a crucial role in the regulation of growth and resistance processes in all kingdoms of life. They have been deeply studied in vertebrates because of their role in drug resistance, but much less is known about ABC superfamily functions in plants.

We have shown that the ABC transporter, multiple drug resistance protein 1 (MDR1, P-glycoprotein) translocates glucosyl ceramide from the cytosolic to the luminal Golgi surface for neutral, but not acidic, gly-cosphingolipid (GSL) synthesis. Here we show that the MDR1 inhibitor, cyclosporin A (CsA) can deplete Gaucher lymphoid cell lines of accumu-lated glucosyl ceramide and Fabry cell lines of globotriaosyl ceramide (Gb3), by preventing de novosynthesis.

The ATP-sensitive potassium (KATP) channel couples glucose metabolism to insulin secretion in pancreatic b-cells. It comprises regulatory sulfonyl-urea receptor 1 and pore-forming Kir6.2 subunits. Binding and⁄or hydro-lysis of Mg-nucleotides at the nucleotide-binding domains of sulfonylurea receptor 1 stimulates channel opening and leads to membrane hyperpolari-zation and inhibition of insulin secretion.

Exposure of the yeastSaccharomyces cerevisiaeto weak organic acids such as the food preservatives sorbate, benzoate and propionate leads to the pronounced induction of the plasma membrane ATP-binding cassette (ABC) transporter, Pdr12p. This protein mediates efflux of weak acid ani-ons, which is essential for stress adaptation.

The human ATP-binding cassette (ABC) transporter, ABCG2 (BCRP⁄ MXR⁄ABCP), is a plasma membrane protein containing intramolecular and intermolecular disulfide bonds and anN-linked glycan at Asn596. We have recently reported that the intramolecular disulfide bond is a critical checkpoint for determining the degradation fates of ABCG2.

The charged quaternary ammonium compounds – methyl, ethyl and benzyl viologens – generate reactive oxygen species in photosynthetic cells. Three independent methyl viologen-resistant spontaneous mutants of Synechocys-tis sp. PCC 6803 were identified, in which the conserved R115 residue of the Slr1174 protein was replaced with G115, L115 and C115. The Slr1174 protein of the DUF990 family is related to the permease subunit of an ABC-2-type transporter and its R115 mutation was found to be solely responsible for the observed methyl viologen resistance. ...

Periplasmic binding proteins are abundant in bacteria by virtue of their essential roles as high-affinity receptors in ABC transport systems and chemotaxis. One of the best studied of these receptors is the so-called glucose⁄galactose-binding protein. Here, we report the X-ray structure of the Salmonella typhimuriumprotein bound to the physiologically relevant ligand, (2R)-glyceryl-b-d-galactopyranoside, solved by molecular replace-ment, and refined to 1.87 A˚ resolution with RandR-free values of 17% and 22%....
