The cytotoxicity of 6 highly cytotoxic Pseudomonas aeruginosa isolates was tested against Vero cells. Cytotoxicity was found to be cell-associated, high cytotoxicity was observed at a multiplicity of infection of 200:1, and this activity gradually decreased as the bacterial count decreased.

Cyclic ADP-ribose (cADPR) is a Ca 2+ mobilizing second messenger found in various cell types, tissues and organisms. Receptor-mediated formation of cADPR may proceed via transmembrane shuttling of the substrate NAD and involvement of the ectoenzyme CD38, or via so far unidentified ADP-ribosyl cyclases located within the cytosol or in internal membranes. cADPR activates intracellular Ca 2+ release via type 2 and 3 ryanodine receptors.

To test the role of poly(ADP-ribose) polymerase on the telomerase activity, we determined the telomerase activity in leukemic cells K562 treated with benzamide and 4-amino 1,8 naphthalimide (NAP), the inhibitors of PARP. We observed that both the agents inhibited telomerase activity in a dose-dependent manner. The doses of benzamide and NAP that inhibited telo-merase activity to 50% of untreated control cells were 10.7 ± 0.6 mmand 200 ± 7lm, respectively.

CD38 is a transmembrane glycoprotein that functions as an ectoenzyme and as a receptor. To assessPseudomonasexotoxin A (ETA) compartmentalization, process-ing and cytotoxicity in vivo, we have studied the fate of internalized ETA with the use of thein vivo rodent liver model following toxin administra-tion, cell-free hepatic endosomes, and pure in vitro protease assays. ETA taken up into rat liver in vivo was rapidly associated with plasma mem-branes (5–30 min), internalized within endosomes (15–60 min), and later translocated into the cytosolic compartment (30–90 min).

Histones are nuclear proteins that package and order the DNA into nucleosomes [1]. Five major families of histones exist: H1 (H5), H2A, H2B, H3, and H4. Two copies of the core histones H2A, H2B, H3 and H4 form the octameric nucleosome core particles [2]. Unlike the other histones, only one copy of the linker histone H1 is present and stabilizes the DNA, which is wrapped around the core nucleosome [3]. Linker histones bind to both the nucleosome and the linker DNA region (approximately 20 to 80 nucleotides in length) between nucleosomes.
