Patients with diabetes mellitus suffer from an increased incidence of complications including cardiovascular disease and cataracts; the mechanisms responsible for this are not fully understood. One characteristic of such complications is an accumulation of advanced glycation end-products formed by the adduction of glucose or species derived from glucose, such as low-molecular mass aldehydes, to proteins.

Laccase-catalyzed polymerization of tyrosine and tyrosine-containing pep-tides was studied in the presence and absence of ferulic acid (FA). Advanced spectroscopic methods such as MALDI-TOF MS, EPR, FTIR microscopy and HPLC-fluorescence, as well as more conventional analyt-ical tools: oxygen consumption measurements and SDS⁄PAGE were used in the reaction mechanism studies. Laccase was found to oxidize tyrosine and tyrosine-containing peptides, with consequent polymerization of the compounds.

Advanced glycation end-products (AGEs) of DNA are formed spontane-ously by the reaction of carbonyl compounds such as sugars, methylglyoxal or dihydroxyacetonein vitro andin vivo. Little is known, however, about the biological consequences of DNA AGEs. In this study, a method was developed to determine the parameters that promote DNA glycation in cultured cells.
