Ammonia oxidizing bacteria

Nitrogen removal with biological methods plays a crucial role in wastewater treatment technology. The treatment begins with the oxidation of ammonia to nitrite to facilitate the subsequent nitrification and denitrification. Various strains of ammonia-oxidising bacteria have been reported. In this study, we use three Bacillus bacteria isolated from swine wastewater to oxidise ammonia.

The present investigation was carried out to understand the effect of ammonia concentrations on the rate of ammonia-oxidizing activity of ammonia oxidizing bacteria (AOB) isolates. Maximum ammonium removal by all the isolates was found at 3 ppm ammonium concentration.

Nitrogen in manure occurs mainly in organic forms (e.g., proteins) and as ammonium (NH4+). Some composted manures contain small amounts of nitrate. Ammonium and nitrate are plant-available forms of N, while organic N is not immediately plant-available. Before organic N can be taken up, however, it must first be converted to inorganic forms. This process, which is completed by soil microbes as a by-product of organic matter decomposition, is called mineralization. The mineralization rate is therefore the rate at which organic N is made plant available.

A lab-scale flat sheet membrane bioreactor (MBR) system was used for the treatment of piggery wastewater to produce an effluent with the appropriate ratio of nitrite:ammonia (1:1 to 1:1.3) as a pre-treatment for the anammox process. The feed wastewater, which was the effluent of a biogas digester, contained 253±49 (n=60) mg.l-1 as COD, 231±18 mg.l-1 as N-ammonia, 223±19 mg.l-1 as total Kjeldalh nitrogen (TKN), alkalinity of 1433±153 mg.l-1 as CaCO3 , and pH=7.5±0.3.

The spatial arrangement of functionally important ammonia oxidizing bacteria (AOB) in activated sludge flocs is largely unknown and activated sludge flocs are regarded as heterogeneous agglomerations of bacterial communities. This study aimed to determine both the location of AOB microcolonies within flocs and the size variation of these microcolonies based on their location.

The plant enzyme phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) shows homology to histidine ammonia-lyase (HAL) whose structure has been solved by X-ray crystallography. Based on amino-acid sequence alignment of the two enzymes, mutagenesis was performed on amino-acid residues that were identical or similar to the active site residues in HAL to gain insight into the importance of this residues in PAL for substrate binding or catalysis. Aspartate ammonia lyases (also referred to as aspartases) catalyze the revers-ible deamination of l-aspartate to yield fumarate and ammonia. In the pro-posed mechanism for these enzymes, an active site base abstracts a proton from C3 ofl-aspartate to form an enzyme-stabilized enediolate intermediate. Ketonization of this intermediate eliminates ammonia and yields the prod-uct, fumarate.
