Nuclear factor erythroid-2 related factor 2 (Nrf2) is a crucial transcription factor that regulates the expression of defensive antioxidants and detoxification enzymes in cells. In a previous study, we showed that expression of the Nrf2 gene is regulated by an epigenetic modification. Rauvolfia verticillata, a traditional Chinese herbal medicine widely used in China, possesses anticancer and antioxidant effects. In this study, we investigated how Nrf2 is epigenetically regulated by reserpine, the main active component in R. verticillata, in mouse skin epidermal JB6 P+ cells.

This study investigated the enzymatic function of two putative plant GPXs, GPXle1 from Lycopersicon esculentum and GPXha2 from Helianthus annuus, which show sequence identities with the mammalian phospholipid hydroperoxide glutathione peroxidase (PHGPX). Both purified recombinant proteins expressed in Escherichia coli show PHGPX activity by reducing alkyl, fatty acid and phospholipid hydroperoxides but not hydrogen peroxide in the presence of glutathione.

Reactive oxygen species (ROS) are products of normal metabolic activities and are thought to be the cause of many diseases. A selenium-containing single-chain abzyme 2F3 (Se-2F3-scFv) that imitates glutathione peroxidase has been produced which has the capacity to remove ROS. To evaluate the antioxidant ability of Se-2F3-scFv, we con-structeda ferrous sulfate/ascorbate (Vc/Fe 2+ )-inducedmito-chondrial damage model system and investigated the capacity of Se-2F3-scFv to protect mitochondria from oxi-dative damage.

Thioredoxins are a group of small redox-active proteins involved in cellular redox regulatory processes as well as antioxidant defense. Thioredoxin, glutaredoxin, and try-paredoxin are members of the thioredoxin superfamily and share structural and functional characteristics. In the mal-arial parasite, Plasmodium falciparum, a functional thio-redoxin and glutathione system have been demonstrated and are considered to be attractive targets for antimalarial drug development.

Leucine zipper-bearing kinase (LZK) is a novel member of themixed lineage kinase (MLK) family [Sakuma, H., Ikeda, A.,Oka,S.,Kozutsumi,Y.,Zanetta,J.P., andKawasaki, T. (1997)J. Biol.Chem.272, 28622–28629].Wehavepreviously shown that LZK activates the c-Jun-NH2terminal kinase (JNK) pathway, but not the extracellular signal-related kinase (ERK) pathway, by acting as a mitogen-activated protein kinase kinase kinase (MAPKKK) [Ikeda, A., Hasegawa, K., Masaki, M., Moriguchi, T., Nishida, E., Kozutsumi, Y., Oka, S., and Kawasaki, T. H2O2 generation is a limiting step in thyroid hormone bio-synthesis. Biochemical studies have confirmed that H2O2is generated by a thyroid Ca 2+ /NADPH-dependent oxidase. Decreased H2O2 availability may be another mechanism of inhibition of thyroperoxidase activity produced by thio-ureylene compounds, as propylthiouracil (PTU) and methimazole (MMI) are antioxidant agents. Therefore, we analyzed whether PTU or MMI could scavenge H2O2or inhibit thyroidNADPHoxidase activityin vitro.

Cys34 in domain I of the three-domain serum protein albumin is the bind-ing site for a wide variety of biologically and clinically important small molecules, provides antioxidant activity, and constitutes the largest portion of free thiol in blood. Analysis of X-ray structures of albumin reveals that the loop containing Tyr84 occurs in multiple conformations. In structures where the loop is well defined, there appears to be an H-bond between the OH of Tyr84 and the sulfur of Cys34.

Glutamine is an essential amino acid in cancer cells and is required for the growth of many other cell types. Glutami-nase activity is positively correlated with malignancy in tumours andwith growth rate in normal cells. In the present work, Ehrlich ascites tumour cells, and their derivative, 0.28AS-2 cells, expressing antisense glutaminase mRNA, were assayed for apoptosis induced by methotrexate and hydrogen peroxide. It is shown that Ehrlich ascites tumour cells, expressing antisense mRNA for glutaminase, contain lower levels of glutathione than normal ascites cells. In addition, 0.

A number of drugs used in cancer chemotherapy induce oxidative stress by generation of oxygen free radicals (ROS) which might be an alternative mechanism for their cytotoxic effect via indu-cing apoptosis. In order to clarify the roles of antioxidants in chemotherapy, we investigated Quercetin (3,3’,4’,5,7-pentahyd-roxyflavone) and N-acetylcysteine (NAC) in different cell types treated with anticancer drugs. We studied cytotoxic activity of Topotecan alone and/or in combination with Quercetin in two human breast cancer cell lines, MCF-7 and MDA-MB-231. ...

Báo cáo khoa học: Dietary antioxidant curcumin inhibits microtubule assembly through tubulin binding
Curcumin, a component of turmeric, has potent antitumor activity against several tumor types. However, its molecular target and mechanism of anti-proliferative activity are not clear. Here, we identified curcumin as a novel antimicrotubule agent. We have examined the effects of curcumin on cellu-lar microtubules and on reconstituted microtubules in vitro.
