Chapter 11 - general transcription factors in eukaryotes. Eukaryotic RNA polymerases, unlike their bacterial counterparts, are incapable of binding by themselves to their respective promoters. Instead, they rely on proteins called transcription factors to show them the way. Such factors are grouped into two classes: general transcription factors and gene-specifi c transcription factors (activators). In this chapter we will survey the general transcription factors that interact with all three RNA polymerases and their promoters.

Cold shock proteins (CSPs) form a family of highly conserved bacterial proteins capable of single-stranded nucleic acid binding. They are suggested to act as RNA chaperones during cold shock inhibiting the formation of RNA secondary structures, which are unfavourable for transcription and translation.

Cyclooxygenase-2 (COX-2) is a key enzyme in the production of prosta-glandins that are major inflammatory agents. COX-2 production is trig-gered by exposure to various cytokines and to bacterial endotoxins. We present here a novel role for the Ets transcription factor ESE-1 in regula-ting the COX-2gene in response to endotoxin and other pro-inflammatory stimuli. We report that the induction of COX-2expression by lipopoly-saccharide (LPS) and pro-inflammatory cytokines correlates with ESE-1 induction in monocyte⁄macrophages. ...

The iron responsive regulator Irr is found in a wide range ofa-proteobac-teria, where it regulates many genes in response to the essential but toxic metal iron. Unlike Fur, the transcriptional regulator that is used for iron homeostasis by almost all other bacterial lineages, Irr does not sense Fe 2+ directly, but, rather, interacts with a physiologically important form of iron, namely heme.
